响应面法优化桑叶黄酮提取工艺及其抗氧化活性研究
Optimization of Extraction Process by Response Sur-face Methodology and Analysis of Antioxidant Ac-tivity of Flavonoid from Mulberry Leaves

Abstract
The objective of this study was to optimize the extraction process of flavonoids from mulberry leaves and explore its antioxidant activity.The experiment selected microwave power,mi-crowave time,ethanol concentration,solid-liquid ratio and extraction time as variables,the yield of flavonoids as the evaluation index.Single factor tests and Box-Behnken method were used to opti-mize the extraction process.The scavenging ability on 1,1-diphenyl-2-picrylhydrazyl(DPPH·),hy-droxyl radical(·OH)and superoxide(O2-·)free radicals of flavonoids from mulberry leaves were also tested to estimate the antioxidant activities in vitro.The results showed that:optimal extraction rate(11.393±0.049 mg/g highest yield)could be achieved under the conditions of microwave power 175 W,microwave time 4.5 min,ethanol concentration 48.5%,solid-liquid ratio 30 mL/g.The abi-lity of mulberry leaves flavonoids extract to scavenge DPPH·,·OH and O2-·was great and scavenge ability was positively correlated with concentration of the extract.To sum up,microwave extraction technology can effectively extract of mulberry leaves flavonoids,leading to better antioxidant acti-vity.关键词
