响应面法优化桑叶黄酮提取工艺及其抗氧化活性研究OA
Optimization of Extraction Process by Response Sur-face Methodology and Analysis of Antioxidant Ac-tivity of Flavonoid from Mulberry Leaves
为了优化桑叶黄酮提取工艺,探讨其抗氧化活性,以微波功率、微波时间、乙醇浓度、液料比和提取次数为影响因素,以桑叶黄酮提取率为考察指标,采用单因素和Box-Behnken法优化提取工艺,并通过测定桑叶黄酮提取物对DPPH·、·OH和O2-·自由基的清除能力,探讨其体外抗氧化活性.结果显示,桑叶黄酮的最佳提取条件为微波功率175 W、微波时间4.5 min、乙醇浓度48.5%、液料比30 mL/g,在此条件下,桑叶黄酮的提取率可达到11.393±0.049 mg/g.桑叶黄酮提取液对DPPH·、·OH和O2-·的清除作用明显,清除能力与黄酮提取液浓度呈正相关关系.综上,应用微波提取技术可有效提取桑叶黄酮,提取的黄酮具有良好的抗氧化活性.
The objective of this study was to optimize the extraction process of flavonoids from mulberry leaves and explore its antioxidant activity.The experiment selected microwave power,mi-crowave time,ethanol concentration,solid-liquid ratio and extraction time as variables,the yield of flavonoids as the evaluation index.Single factor tests and Box-Behnken method were used to opti-mize the extraction process.The scavenging ability on 1,1-diphenyl-2-picrylhydrazyl(DPPH·),hy-droxyl radical(·OH)and superoxide(O2-·)free radicals of flavonoids from mulberry leaves were also tested to estimate the antioxidant activities in vitro.The results showed that:optimal extraction rate(11.393±0.049 mg/g highest yield)could be achieved under the conditions of microwave power 175 W,microwave time 4.5 min,ethanol concentration 48.5%,solid-liquid ratio 30 mL/g.The abi-lity of mulberry leaves flavonoids extract to scavenge DPPH·,·OH and O2-·was great and scavenge ability was positively correlated with concentration of the extract.To sum up,microwave extraction technology can effectively extract of mulberry leaves flavonoids,leading to better antioxidant acti-vity.
