Abstract

[Objective]The aim of this study was to investigate the multidrug-resistant characteristics of blaCMY-2-positive Proteus Mirabilis and analyze the whole-genome sequence structure of the strain CY32.[Method]Five blaCMY-2-positive P.mirabilis strains were tested for florfenicol resistance and plasmid-mediated fluoroquinolone resistance genes,conjugation experiments,and Southern hybridiza-tion of the blaCMY2 gene.Whole genome sequencing and bioinformatics analysis were carried out on Proteus Mirabilis strain CY32.[Result]The result of Southern hybridization revealed blaCMY-2 gene was located in chromosomes of five strains of Proteus Mirabilis.The floR was detected in four strains(CY12、CY32、S31 and S52),and qnrD was detected in two strains(CY12 and CY32).The se-quence analysis demonstrated CY32 contained both SXT/R391 ICEs(ICEPmiJpn1)and PmGRI1 re-sistance genomic islands in chromosome.Two composite transposons bracketed by IS10 were located in variable region V of ICEs in a tandem array,and one of which carried the blaCMY2.The PmGRI1 drug resistance island of CY32 contained 12 resistance genes.Compared with PmGRI1 in other Proteus Mir-abilis strains,the multi-drug resistance region with the largest difference was located in the Tn21 trans-poson.In addition,CY32 harbored two plasmids,including afloR-carrying IncQ plasmid and a qnrD-carrying non-transmissible plasmid,belonged to an undescribed incompatibility group.The 15 antimi-crobial resistance genes were carried in CY32 in total,exhibiting the characteristics of multidrug resist-ance.[Conclusion]Acquisition of multidrug resistance in Proteus Mirabilis through genomic islands and plasmids would make it more difficult to treat infection caused by Proteus Mirabilis,suggesting that the monitoring of such resistant Proteus Mirabilis should be strengthened.
