Optimization of ultrasound-assisted cellulase extraction of selenium polysaccharide from wheat and its antioxidant activity

摘要

Abstract

Selenium polysaccharides were extracted from selenium-rich whole wheat flour using ultrasound-as-sisted cellulase method.The extraction process of wheat selenium polysaccharides was optimized by single-fac-tor experiment and response surface methodology,with the extraction rate of selenium polysaccharides as the e-valuation index.The antioxidant activity of wheat selenium polysaccharides against ABTS+ was also studied.The results showed that the optimal extraction conditions for wheat selenium polysaccharides were a solid-liquid ratio of 1:17(g:mL),cellulase concentration of 0.75%,ultrasound temperature of 60℃,ultrasound power of 497 W,and ultrasound time of 50 minutes.Under these conditions,the extraction rate of wheat selenium pol-ysaccharides was38.24%,and the selenium content was 0.093 mg/kg.The vitro antioxidant activity study showed that the IC50 value of wheat selenium polysaccharides for ABTS+ radical scavenging was4.56 mg/mL.
