HLA-G 3'UTR基因多态性及血清可溶性HLA-G水平与儿童诺如病毒感染的相关性研究OACSTPCD
Association of HLA-G 3'UTR gene polymorphism and serum soluble HLA-G level with norovirus infection in children
目的 探讨人类白细胞抗原-G(HLA-G)3'非翻译区(3'UTR)基因多态性(SNP)及血清可溶性HLA-G(sHLA-G)水平与儿童诺如病毒感染的相关性.方法 选取2021年1月至2022年12月新乡医学院第一附属医院收治的346例诺如病毒感染患儿作为病例组,另选取同期320例健康体检儿童作为对照组,留取所有研究对象全血标本并提取DNA.采用聚合酶链反应扩增 HLA-G 3'UTR基因,产物外送测序.使用SNPstats在线分析软件计算两组间SNP位点基因型和等位基因分布频率.采用酶联免疫吸附试验检测病例组和对照组血清sHLA-G水平.结果 病例组和对照组HLA-G 3'UTR 14 bp+/-和+3142 C/G基因型和等位基因分布频率比较,差异均无统计学意义(P>0.05).病例组血清sHLA-G水平明显高于对照组,差异有统计学意义(P<0.05);诺如病毒不同程度感染组血清sHLA-G水平比较,差异均无统计学意义(P>0.05).病例组和对照组14 bp+/-基因型和+3142 C/G基因型对应的血清sHLA-G水平比较,差异均无统计学意义(P>0.05).结论 HLA-G 3'UTR SNP与诺如病毒感染的易感性无关.血清sHLA-G水平对诺如病毒感染早期诊断有较好的预测价值,可能是一种新的血清生物标志物,与患者病情严重程度及预后的关联尚需进一步验证.
Objective To investigate the association of human leukocyte antigen-G(HLA-G)3'untranslat-ed region(3'UTR)gene polymorphism(SNP)and serum soluble HLA-G(sHLA-G)levels with norovirus infection in children.Methods A total of 346 children with norovirus infection admitted to the First Affiliated Hospital of Xinxiang Medical University from January 2021 to December 2022 were selected as the case group,and 320 healthy children who underwent physical examination during the same period were selected as the control group.Whole blood samples of all subjects were collected and DNA was extracted.HLA-G 3'UTR gene was amplified by polymerase chain reaction(PCR)and the products were sent out for sequencing.SNPstats online analysis software was used to calculate the genotype and allele frequency distribution of SNP sites between the two groups.Enzyme-linked immunosorbent assay was used to detect serum sHLA-G levels in case group and control group.Results There was no significant difference in HLA-G 3'UTR 14 bp +/-and +3142 C/G genotypes and allele frequencies between the case group and the control group(P>0.05).The serum level of sHLA-G in the case group was significantly higher than that in the control group(P<0.05).There was no significant difference in serum sHLA-G levels between groups with different degrees of no-rovirus infection(P>0.05).There was no significant difference in serum sHLA-G levels corresponding to 14 bp+/-genotype and +3142 C/G genotype between case group and control group(P>0.05).Conclusion HLA-G 3'UTR SNP is not associated with susceptibility to norovirus infection.Serum sHLA-G level has a good predic-tive value for the early diagnosis of norovirus infection,and may be a new serum biomarker.The association with the severity and prognosis of patients needs to be further verified.
