Effect of GABA-expressing neurons in locus coeruleus on body tempera-ture

Abstract
AIM:The locus coeruleus plays a crucial role in energy homeostasis,notably through its γ-amino-butyric acid(GABA)neurons involved in body temperature regulation.However,the specific function of locus coeruleus GABA-expressing neurons in regulating body temperature is not fully understood.This study aims to investigate the role of locus coeruleus GABA-expressing neurons in thermoregulation and the underlying mechanisms.METHODS:Chemoge-netic techniques were employed to selectively inhibit GABAergic neuron activity in the locus coeruleus.Subsequent obser-vations included changes in core body temperature,food intake,glucose tolerance,and insulin sensitivity in mice.Fur-thermore,the expression of mitochondrial uncoupling protein 1(UCP1)in interscapular brown adipose tissue(IBAT)was detected by Western blot.RESULTS:The inhibition of locus coeruleus GABA-expressing neurons led to a significant re-duction in core body temperature and IBAT temperature in fasted mice(P<0.05),along with a marked increase in UCP1 expression in IBAT(P<0.05).CONCLUSION:The findings of this study reveal the significant influence of locus coeru-leus GABA-expressing neurons on body temperature regulation in mice,advancing the understanding of how the central nervous system impacts thermoregulation and energy metabolism by modulating specific neuronal groups.This contributes to the broader comprehension of related physiological mechanisms.关键词
