目的:蓝斑核作为能量调节枢纽参与调节能量稳态,γ-氨基丁酸(GABA)神经元发挥体温调节作用,然而蓝斑核GABA表达神经元在体温调节方面的作用尚未明确.基于此,本研究旨在探索蓝斑核GABA表达神经元是否参与体温调节.方法:运用化学遗传方法特异性抑制小鼠蓝斑核GABA能神经元活性,统计其体温、采食量、糖耐量、胰岛素耐受性变化.同时,通过Western blot检测肩胛间棕色脂肪组织(IBAT)中线粒体解偶联蛋白1(UCP1)的表达.结果:抑制蓝斑核GABA表达神经元导致饥饿状态小鼠核心体温和IBAT温度显著下降(P<0.05),同时IBAT中的UCP1表达水平显著上升(P<0.05).结论:本研究结果表明蓝斑核GABA表达神经元可调控小鼠体温,为中枢神经系统如何通过调节特定神经元群体来影响体温和能量代谢提供了新的视角,这对于理解相关的生理机制具有重要意义.

AIM:The locus coeruleus plays a crucial role in energy homeostasis,notably through its γ-amino-butyric acid(GABA)neurons involved in body temperature regulation.However,the specific function of locus coeruleus GABA-expressing neurons in regulating body temperature is not fully understood.This study aims to investigate the role of locus coeruleus GABA-expressing neurons in thermoregulation and the underlying mechanisms.METHODS:Chemoge-netic techniques were employed to selectively inhibit GABAergic neuron activity in the locus coeruleus.Subsequent obser-vations included changes in core body temperature,food intake,glucose tolerance,and insulin sensitivity in mice.Fur-thermore,the expression of mitochondrial uncoupling protein 1(UCP1)in interscapular brown adipose tissue(IBAT)was detected by Western blot.RESULTS:The inhibition of locus coeruleus GABA-expressing neurons led to a significant re-duction in core body temperature and IBAT temperature in fasted mice(P<0.05),along with a marked increase in UCP1 expression in IBAT(P<0.05).CONCLUSION:The findings of this study reveal the significant influence of locus coeru-leus GABA-expressing neurons on body temperature regulation in mice,advancing the understanding of how the central nervous system impacts thermoregulation and energy metabolism by modulating specific neuronal groups.This contributes to the broader comprehension of related physiological mechanisms.
