目的 检测琥珀酸脱氢酶A(SDHA)在胃癌组织及细胞中的表达水平,探讨其对胃癌增殖、迁移和侵袭能力的影响.方法 通过免疫组织化学法检测SDHA在胃癌组织及癌旁组织中的表达情况.结合Western blotting检测SDHA在正常胃上皮细胞与胃癌细胞中的表达.通过Transwell实验、划痕实验、CCK-8实验、克隆形成实验进一步研究异常表达SDHA对胃癌细胞迁移、侵袭和增殖能力的影响.结果 免疫组织化学及Western blotting结果提示,SDHA在胃癌组织及胃癌细胞中高表达.敲低及过表达SDHA可影响胃癌细胞的增殖、迁移和侵袭.此外,SDHA表达与肿瘤免疫浸润显著相关.结论 SDHA能够促进胃癌细胞增殖、迁移和侵袭,有望成为胃癌治疗的潜在靶点.

Objective To explore the effect of succinate dehydrogenase A(SDHA)on the biological characteristics of gastric cancer cells by detecting its expression level in gastric cancer tissues and cells.Methods The expression of SDHA in gastric cancer and para-neoplastic tissues was analyzed using immunohistochemistry(IHC).Combined with Western blotting to detect SDHA expression in normal gastric epithelial and gastric cancer cells,the Transwell,scratch,Cell Counting Kit-8(CCK-8),and colony formation assays were used to further investigate the effects of abnormal SDHA expression on the migration,invasion,and proliferation ability of gastric cancer cells.Results IHC and Western blotting results showed that SDHA was highly expressed in gastric cancer tissues and cells.Knockdown and overexpression of SDHAaffected the proliferation,migration,and invasion abilities of gastric cancer cells.In addition,SDHA expression was significantly correlated with tumor immune infiltration.Conclusion SDHA affects the prognosis of patients with gastric cancer,alters the biological functions of gastric cancer cells,and is therefore a potential target for gastric cancer therapy.
