Research progress of STING agonists
cGAS-STING通路作为固有免疫系统的重要组成部分,可以识别来自病原微生物和宿主细胞的胞质DNA,诱导Ⅰ型干扰素的分泌和炎症细胞因子的生成,激活机体免疫系统,抵抗病原微生物的入侵及增强机体抗肿瘤活性等.该文简要介绍了 cGAS-STING通路的生物学功能,主要聚焦于STING激动剂的研究进展,包括CDN类和Non-CDN类STING激动剂的药理作用及其研究阶段,总结与展望STING激动剂研发过程中的问题与挑战,以期为后续相关药物的研发和疾病治疗提供思路.
As an important part of the innate immune system,the cGAS-STING pathway can recognize cytoplasmic DNA from pathogenic microorganisms and host cells,induce the secretion of type Ⅰ interferon and the production of inflammatory cytokines,activate the immune system,resist the invasion of pathogenic mi-croorganisms,and enhance antitumor activity.Here we briefly introduce the mechanism and biological functions of the cGAS-STING pathway,focus on the current research progress of STING agonists,including the pharmacological effects and research sta-ges of STING CDN and Non-CDN agonists,and finally summari-zes and prospects the problems and challenges in the research of STING agonists,in order to provide ideas for the development of related drugs and disease treatment.
