The regulatory mechanism of ferroptosis in bone metabolism
铁死亡是一种新型的细胞死亡方式,以细胞内铁过载和脂质过氧化物过度积累为特征,与多种骨科疾病的发生密切相关.骨质疏松是以骨密度降低为主要病理表现的慢性代谢性骨病,近年来随着研究的不断深入,发现铁死亡对骨代谢着重要的影响,而通过对铁死亡的调节以维持骨稳态,在骨质疏松的防治领域取得了一定的突破,展现出了良好的应用前景.该文阐述了铁死亡的发生机制和调节途径,梳理了铁死亡对骨代谢的调控机制,并总结了铁死亡在骨质疏松症发病及治疗中的研究进展,以期拓宽骨代谢的研究思路.
Ferroptosis is a novel mode of cell death characterized by intracellular iron overload and excessive accumulation of lipid peroxides,which is closely associated with the pathogenesis of many orthopedic diseases.Osteoporosis is a chronic metabolic bone disease characterized by decreased bone mineral density.In recent years,a large number of studies have been conducted to regulate bone homeostasis through ferroptosis to treat osteopo-rosis,and certain results have been obtained,showing a good research prospect.This article summarizes the mechanism and regulation pathway of ferroptosis,explores the correlation be-tween ferroptosis and the pathological mechanism of osteoporo-sis,systematically elaborates on the effect of ferroptosis in osteo-blasts,osteoclasts and bone marrow mesenchymal stem cells,and sorts out the therapeutic application of ferroptosis for osteo-porosis in order to expand the therapeutic ideas of osteoporosis.
