摘要

Abstract

Aim To investigate the critical effects of sulfated polysaccharide from Undaria pinnatifida(SP-UP)on hepatocellular carcinoma(HCC)cells and its mechanisms.Methods MTT assay and colony forma-tion assay were used to verify the effect of SPUP on the proliferation of HepG2 and SMMC-7721 cells.The ap-optotic effect of SPUP on HepG2 and SMMC-7721 cells was detected by Hoechst 33258 and Western blot.The metastasis and invasion of HCC cells were detected by Transwell and scratch methods.The autophagy,epi-thelial-mesenchymal transition(EMT),matrix metal-loproteinase 2(MMP-2)and MMP-9 levels in HCC cells were investigated by immunofluorescence and Western blot.Results SPUP had short-term and long-term inhibitory effects on HepG2 and SMMC-7721 cells,minimal inhibitory effect on L-02 cells,and SP-UP could promote apoptosis and autophagy of HCC cells.Meanwhile,SPUP could suppress migration and invasion abilities,EMT and MMPs levels of HCC cells.Conclusions SPUP inhibits the growth and mi-gration of HCC cells via promoting apoptosis,autoph-agy,and inhibiting EMT and MMPs.
