Effects of Paeonia rockii pollen on D-galactose aging model mice

Abstract
Aim To investigate the protective effect of P.rockii pollen on the improvement of learning memo-ry and oxidative stress damage in D-galactose-induced aging mice.Methods Sixty C57BL/6 mice were ran-domly divided into blank group,model group,positive group,low-,medium-,and high-dose groups(750,1 500,3 000 mg·kg-1)of P.rockii pollen,and D-galactose(500 mg·kg-1)was injected subcu-taneously on the back of the neck to replicate the suba-cute aging model.Morris water maze was used to eval-uate the learning and memory of mice,HE and DAPI staining were used to evaluate the damage of liver and brain tissue cells,and ELISA was used to detect the levels of MDA,T-AOC,GSH-Px,SOD,CAT and γ-H2AX and SA-β-Gal in serum,liver and brain tis-sues.Results The water maze experiment showed that P.rockii pollen could significantly improve the learning and memory of aging mice.Tissue staining showed that P.rockii pollen had a protective effect on liver and brain tissue cell injury in D-galactose-induced aging mice,and the effect was most obvious in the high-dose group.ELISA results showed that compared with the model group,the levels of MDA,γ-H2AX and SA-β-Gal in each dose group of P.rockii pollen decreased,and the levels of T-AOC,GSH-Px,SOD and CAT increased.Conclusions P.rockii pollen can improve the learning and memory of D-galactose-induced aging model mice,improve the activity of an-tioxidant enzymes and the ability to scavenge free radi-cals and lipid peroxides in aging mice,and protect liv-er and brain tissue cells from oxidative damage,sug-gesting that it has a potential anti-aging effect.关键词
