基于TCGA数据挖掘及网络药理学探讨卤泛群治疗胶质母细胞瘤的潜在机制
The potential mechanism of halogenated pangroup treatment for glioblastoma based on TCGA data mining and network pharmacology

Abstract
Aim To explore the potential of the drug halofantrine for treating glioblastoma,utilizing the canc-er genome atlas(TCGA)genomic data and further il-lustrating its mechanism through network pharmacolo-gy.Methods Key genes associated with the develop-ment of glioblastoma were identified from the open cancer clinical database TCGA and the potential drugs for glioblastoma were searched via the CMAP database.Drug target information was collected from STITCH,Drugbank,BindingDB,Batmant-Tcm,TargetNet,Swiss Target,and PubChem.Visualization network diagrams of"Drug,active ingredient,and target"were then con-structed through Cytoscape,with the mechanism of ac-tion predicted by GO,KEGG,and molecular docking a-nalysis.The effects of halofantrine on glioblastoma cells and therapeutic targets were tested through CCK8 and qPCR.Results Halofantrine was identified as the potential drug for treating glioblastoma,with 150 targets,173 glioblastoma therapeutic targets,and 12 in-tersection targets.The three core targets were identi-fied:EGFR,MAPK14,and IGF1R.Molecular docking analysis showed that halofantrine had good binding po-tential to the core targets.Conclusions Halofantrine plays a role in the treatment of glioblastoma through its action on EGFR,MAPK14,and IGF1R core targets.关键词
