目的 建立UPLC-Q-TOF-MS/MS法分析紫辛鼻鼽颗粒化学成分及入血成分.方法 分析采用Agilent ZORBAX SB-C18色谱柱(1.8 μm,2.1 mm×100 mm);流动相 0.2%甲酸-乙腈,梯度洗脱;体积流量 0.2 mL/min;柱温 30℃;电喷雾离子源;正负离子扫描.根据准分子离子、保留时间、二级碎片离子结合对照品信息及文献报道进行成分鉴定.结果 共鉴定出 88 种成分,其中 28 种经对照品比对确认.从大鼠血清中发现了 30 种成分,其中 20 种为原型成分,10 种为代谢产物.结论 该方法稳定可靠,可为紫辛鼻鼽颗粒药效物质基础、质量控制研究奠定基础.

AIM To establish a UPLC-Q-TOF-MS/MS method for the analyses of chemical constituents and constituents absorbed into blood in Zixin Biqiu Granules.METHODS The analysis was performed on a 30℃thermostatic Agilent ZORBAX SB-C18 column(1.8 μm,2.1 mm×100 mm),with the mobile phase comprising of 0.2%formic acid-acetonitrile flowing at 0.2 mL/min in a gradient elution manner,and electron spray ionization source was adopted in positive and negative ion scanning with multiple reaction monitoring mode,after which constituent identification was employed according to excimer ions,retention time,secondary fragment ions combined with reference substance information,relevant literatures.RESULTS Total 88 constituents were identified,28 of which were validated by comparison of reference substances.Thirty constituents were observable in rat serum,20 of which were prototype constituents,10 of which were metabolites.CONCLUSION This stable and reliable method can lay the foundation for research on pharmacological substance basis and quality control of Zixin Biqiu Granules.
