目的:采用超高效液相色谱-质谱法(UPLC-MS/MS)建立心悦胶囊中主要成分人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1、拟人参皂苷F11、人参皂苷Rd和 20(S)-人参皂苷F1 的含量测定方法.方法:使用Waters ACQUITY BEH C18 色谱柱(100 mm×2.1 mm,1.7 μm),以乙腈-水为流动相,梯度洗脱,流速为 0.5 mL·min-1,柱温为 40℃,电喷雾电离源,多反应离子监测扫描方式进行检测.结果:6 个成分分别在质量浓度为 9.638~963.800、9.839~983.900、9.951~995.100、5.270~1 054.000、9.608~960.800、4.905~981.000 ng·mL-1 时与峰面积线性关系良好(r>0.998),平均加样回收率(RSD)分别为 96.9%(2.2%)、100.1%(1.5%)、97.2%(1.8%)、98.5%(2.3%)、96.6%(2.8%)、100.2%(3.5%).结论:所建立的方法快速、准确、灵敏度高,可用于心悦胶囊中西洋参茎叶总皂苷的质量控制.

Objective:To establish the method for determination of ginsenoside Rg1,ginsenoside Re,ginsenoside Rb1,pseudo-ginsenoside F11,ginsenoside Rd and 20(S)-ginsenoside F1 in Xinyue Capsules by ultra-high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).Methods:The analysis was performed on Waters ACQUITY BEH C18 column(100 mm×2.1 mm,1.7 μm),with acetonitrile(A)-water(B)as the mobile phase,with gradient elution at a flow rate of 0.5 mL·min-1.The column temperature was set at 40℃.The analytes were detected using the electrospray ionization(ESI)source in multiple reaction monitoring(MRM)scan mode.Results:Six constituents showed good linear relationships within the concentration ranges of 9.638-963.800,9.839-983.900,9.951-995.100,5.270-1 054.000,9.608-960.800,4.905-981.000 ng·mL-1(r>0.998),whose average recoveries(RSD)were 96.9%(2.2%),100.1%(1.5%),97.2%(1.8%),98.5%(2.3%),96.6%(2.8%),100.2%(3.5%),respectively.Conclusion:The established method is rapid,accurate,and highly sensitive,making which can be used for quality control of total ginsenosides from Panax quinquefolium stems and leaves in Xinyue Capsules.
