血府逐瘀汤对人食管癌ECA-109荷瘤裸鼠肿瘤血管的影响
Effect of Xuefu Zhuyu Decoction on Tumor Blood Vessels of Esophageal Cancer ECA-109 Tumor Bearing Nude Mice

Abstract
Objective:To observe the effect of Xuefu Zhuyu Decoction(XFZYD)on the vessel of tumor tissue in nude mice with tumors.Methods:A nude mouse model with subcutaneously transplanted esophageal cancer was constructed using human esophageal cancer cells ECA-109.After being modeled,forty-five nude mice were randomly divided into a control group and low-dose and high-dose groups of XFZYD,with 15 mice in each group.The control group was gavaged with 0.9%sodium chloride solution,and the low and high dose XFZYD groups were gavaged with 1.2 g/mL and 4.8 g/mL of XFZYD.Tumor tissues were removed after 14 d of continuous intervention.Tumor volume and tumor suppression rate were calculated.Tumor vascular density,α-SMA positive cell coverage,Collegen-Ⅳ coverage,CD31 positive rate and vascular leakage rate were detected by immunofluorescence assay.The oxygen-depleted area of tumor tissue was detected by immunohistochemistry.Results:The tumor inhibition rate,microvessel density,α-SMA positive cell coverage rate and Collegen-Ⅳ coverage rate of Xuefu Zhuyu Decoction in high and low dose groups were significantly improved compared with the control group(P<0.01,P<0.05),the vascular leakage rate and the area ratio of tumor oxygen-depleted area were lower than those in the control group(P<0.01).Conclusion:Xuefu Zhuyu Decoction can inhibit the growth of subcutaneous transplanted tumor in mouse with the tumor transplanted from human esophageal carcinoma,improve the function and structure of tumor blood vessels and the hypoxia state of tumor tissue,and promote the normalization of tumor blood vessels to a certain extent.关键词
