体外抗猪轮状病毒8种藏药的初步筛选OA北大核心CSTPCD
Preliminary Screening of 8 Tibetan Medicines against Porcine Rotavirus in vitro
为筛选具有抗病毒作用的藏药,对前期研究发现具有潜在抗病毒作用的诃子、甘青乌头、矮紫堇、甘青青兰、川西獐牙菜、短穗兔耳草、铁棒锤、瑞香狼毒等8种藏药的提取物进行了体外抗病毒研究.用水提法对8种藏药进行处理,通过浓缩、真空干燥后配制成不同浓度的水提物,以猪轮状病毒(PoRV)为受试对象,利巴韦林为对照.采用CCK-8测定药物安全浓度,用细胞病变效应(CPE)观察、实时荧光定量PCR、间接免疫荧光相结合的方法测定8种药物体外抗猪轮状病毒作用.结果表明,诃子、甘青乌头、甘青青兰、短穗兔耳草、川西獐牙菜、铁棒锤、瑞香狼毒、矮紫堇8种藏药提取物和利巴韦林药物对MA-104细胞的最大安全浓度分别为0.02、0.49、0.98、2.60、7.81、15.63、1.30、31.25、0.20 mg/mL.在药物与病毒预先作用的方式下,川西獐牙菜、瑞香狼毒、短穗兔耳草、铁棒锤、诃子5种藏药提取物和利巴韦林对猪轮状病毒的最大抑制率均超过50%,分别为87.37%、91.55%、86.19%、91.88%、63.74%、96.33%,能有效减轻病毒感染所致的细胞病变效应,其余3种藏药提取物对PoRV致细胞病变的最大抑制率均低于20%.对PoRV最大抑制率超过50%的5种藏药进一步试验发现,5种藏药提取物显著降低了病毒的复制增殖能力(P<0.001),有效减少了病毒在MA-104细胞的感染.研究结果证明5种藏药提取物对PoRV具有一定的抗病毒作用,为抗病毒藏药的筛选和机制研究提供了基础资料.
In order to screen Tibetan medicines with antiviral effects,in vitro antiviral studies are carried out on extracts of eight Tibetan medicines,including Terminalia chebula,Aconitum tanguticum,Corydalis edulis,Dracocephalum tanguticum,Swertia mussotii,Lagotis brachystachya,Aconitum pendulum,Stellera chamaejasme,which are found to have potential antiviral effects.Eight Tibetan medicines were extracted with water.Water extracts of different concentrations were prepared by concentrating and vacu-um drying.Porcine rotavirus(PoRV)was used as the test object.The safe concentration of the medicines was determined by CCK-8 and the anti-porcine rotavirus effect of eight medicines in vitro was determined by the combination of cytopathic effect(CPE)observation,real-time quantitative PCR and indirect immunofluorescence assay.At the same time,ribavirin was used as an antiviral control.The result showed that the maximum safe concentrations of eight Tibetan medicinal extracts of Terminalia chebula,Aconitum tanguticum,Dracocephalum tanguticum,Lagotis brachystachya,Swertia mussotii,Aconitum pendulum,Stellera chamaejasme,Corydalis edulis and ribavirin were 0.015 mg/mL,0.49 mg/mL,0.98 mg/mL,2.60 mg/mL,7.81 mg/mL,15.63 mg/mL,1.30 mg/mL,31.25 mg/mL and 0.20 mg/mL,respectively.Antiviral tests showed that after the medicinal interactions with the viruses,the maximum inhibitory rates of Swertia mussotii,Stellera chamaejasme,Lagotis brachystachya,Aconitum pendulum,Terminalia chebula and ribavirin against porcine rotavirus were all over 50%,which were 87.37%,91.55%,86.19%,91.88%,63.74%and 96.33%,respectively.It could effectively reduce the cytopathic effect caused by viral infection.The maximum inhibitory rates of the other three extracts on PoRV cyto-pathic effect was lower than 20%.Further tests on the five Tibetan medicines with the maximum inhibition rate of more than 50%of PoRV showed that the five Tibetan medicine extracts significantly reduced the a-bility of virus replication(P<0.05)and effectively reduced the infections of virus in MA-104 cells.This study revealed that five extracts of Tibetan medicines have certain antiviral effects on PoRV,which pro-vides basic data for screening and mechanism research of antiviral Tibetan medicines.
