洛那法尼对动物源丁型肝炎病毒作用的初步研究
Possible Effect of Lonafarnib on Animal-derived Hepatitis D Like Virus

Abstract
The aim of the study is to investigate the effect of Lonafarnib(LNF)on the animal-derived hepatitis D virus(HDV),which will provide insights of animal-derived HDV like virus.We modified HDV infectious clone to recapitulate the key features of animal HDV like virus by using site-directed mutagenesis(SDM),followed by transfection and antibiotic screening.Virus replication and HDAg expression were detected by Western blot and indirect immunofluorescence assay.In addition,the effect of LNF on animal HDV replication was detected by real-time quantitative PCR.The results showed that the modified HDV clones can replicate efficiently in vitro,representing a proper model to study animal HDV like virus.Compared with the untreated group,LNF treatment increased viral RNA levels in HDV model,while no significant effect on the modified HDV clones.This implies that the life cycle and drug responses of animal-derived HDV like viruses are distinct compared with HDV.关键词
