丹黄散对糖尿病小鼠皮肤创面愈合的促进作用及调节细胞自噬机制OACSTPCD
Promoting effect of Danhuang powder on skin wound healing in diabetic mice and its mechanism of regulating autophagy
目的:探讨丹黄散对糖尿病小鼠溃疡创面愈合过程中自噬相关蛋白管连接蛋白轻链 3(LC3)及苄氯素 1(Beclin1)表达的影响.方法:18 只自发 2 型糖尿病 db/db小鼠随机分为模型组、生长因子组、丹黄散组,空白组为 6只 db/m小鼠,制备糖尿病创面模型后空白组、模型组仅清创,不予药物处理,生长因子组、丹黄散组清创后予以相应药物外敷,每日 1 次,连续干预 21d.造模后第 7、14、21 天观察比较创面愈合情况、HE染色观察创面病理情况、透射电镜下组织线粒体与自噬小体情况、免疫组化法检测创面组织 LC3/Beclin1 的表达.结果:观察第 7、14、21 天空白组、生长因子组、丹黄散组创面愈合率均较模型组高(P<0.05);HE染色结果,丹黄散组与模型组、空白组相比较,丹黄散组新生血管和成纤维细胞增多、炎性细胞浸润较轻;透射电镜下发现,与模型组、空白组相比较,生长因子组和丹黄散组创面胞浆中出现较多自噬小体;免疫组化法检测发现空白组、生长因子组、丹黄散组小鼠创面组织 LC3、Beclin1 蛋白表达增强.结论:丹黄散可能通过上调组织中LC3/Beclin1 表达,达到改善小鼠溃疡创面愈合效果.
Objective:To investigate the effect of Danhuang powder on the expression of autophagy protein LC3/Beclin1 during ulcer wound healing in diabetic mice.Methods:18 db/db mice with spontaneous type 2 diabetes were randomly divided into model group,growth factor group and Danhuang powder group,and 6 db/m mice were included in the blank group and model group.After the diabetes wound model was prepared,the blank group and model group were only debrideed without drug treatment,and the growth factor group and Danhuang powder group were treated with the corresponding drug after debridement,once a day for 21 days.Wound healing,wound pathology with HE staining,mitochondria and autophagosomes under transmission electron microscopy,and LC3/Beclin1 ex-pression in wound tissue were detected by immunohistochemical method at 7,14 and 21 days after the molding.Results:The wound healing rate of 7,14,21 days blank group,growth factor group and Danhuang powder group was higher than that of model group(P<0.05).HE staining results showed that compared with model group and blank group,Danhuang powder group had higher increase of neovascularization and fibroblasts,the expression of LC3 and Beclin1 protein in wound tissue of mice in blank group,growth factor group and Danhuang powder group was en-hanced by immunohistochemistry.Conclusion:Danhuang powder may improve the healing effect of ulcer wound in mice by up-regulating LC3/Beclin1 expression.
