Comparative study on the Dynamic Characteristics of PEDV G2 mutant strain

Abstract
[Objective]This study aimed to comprehensively investigate the characteristics of a porcine epidemic diarrhea virus(PEDV)strain(PEDV-BJ-150)after continuous passages,exploring its biological properties,genetic variations,pathogenici-ty,and cellular adaptability,to evaluate its potential as a candidate attenuated vaccine strain.[Methods]Cellular culture,ani-mal infection experiments,and genome sequencing were conducted on PEDV-BJ-150 and the original PEDV-BJ-01 strain to evaluate their proliferation capability,viral titers,pathogenicity in different cell lines,nucleotide sequence variations,and to as-sess infection characteristics and pathological changes using an animal model.[Results]After continuous passaging,PEDV-BJ-150 exhibited enhanced proliferation ability and titers in Vero-M cells,particularly demonstrating significant titer differences be-tween suspension-adapted and adherent Vero-M strains.Experimental findings showed reduced pathogenicity in vivo post-infec-tion,especially with undetectable virus presence in the lungs,while higher viral titers were observed in nasal turbinates and small intestines.Additionally,genome sequencing revealed multiple amino acid mutations in the S protein of PEDV-BJ-150,potentially influencing its cellular tropism and pathogenicity.[Conclusion]This study revealed a significant decrease in pathoge-nicity and virulence of PEDV-BJ-150 after continuous passages,indicating its potential as an attenuated vaccine strain.Specifi-cally,continuous amino acid deletions in the S protein may contribute to reduced viral pathogenicity,offering new insights for vaccine development.Moreover,the study identified tissue-specific differences in PEDV infection characteristics,which are crucial for understanding virus transmission mechanism.The innovation of this study lies in discovering changes in pathogenici-ty post-continuous passages of PEDV and the impact of S protein mutations on viral characteristics,providing new insights for virus evolution and vaccine design.This research offers critical clues for understanding the virological characteristics and viru-lence changes of PEDV,holding potential implications for virus vaccine development and outbreak control.关键词
