清热养阴除湿汤对类风湿关节炎模型大鼠肠道菌群的影响
Effects of Qingre Yangyin Chushi Decoction on Intestinal Flora in Rheumatoid Arthritis Model Rats
摘要
Abstract
Objective To observe the effects of Qingre Yangyin Chushi Decoction on inflammation factors and intestinal flora of rheumatoid arthritis(RA)model rats.Methods The RA model rats were established through collagen.The model rats were randomly divided into model group,positive drug group,and TCM group,and a normal group was set up,with 6 rats in each group.The administration groups were given corresponding drugs by gavage for 5 weeks.The histopathological changes of ankle joint were observed by HE staining;the serum inflammatory factors TGF-β,IFN-γ,IL-4,IL-17A contents were detected by ELISA;16S rRNA technology and Illumina high-throughput gene sequencing platform were used to sequence and analyze of the structural composition and community differences of intestinal flora.Results Compared with the normal group,the model group rats had local disappearance of ankle joint cartilage,decreased and narrowed bone trabeculae,significantly thickened synovium,and a large amount of inflammatory cell infiltration,serum pro-inflammatory factor IFN-γ and IL-17A contents increased(P<0.01),anti-inflammatory factor TGF-β and IL-4 contents decreased(P<0.01).Compared with the model group,inflammatory cell infiltration and cartilage surface erosion were reduced in the ankle joint of rats in the TCM group,serum IFN-γ and IL-17A contents were reduced,and TGF-β and IL-4 contents increased(P<0.01).The sequencing analysis of intestinal flora showed that Qingre Yangyin Chushi Decoction could regulate the composition and abundance of intestinal flora in model rats,including decreasing the abundance of Odoribacter,Christensenella,and Povalibacter(P<0.05),increasing the abundance of Arthrobacter,Corynebacterium,Ralstonia,Enterorhabdus and Parvibacter(P<0.05).Conclusion Qingre Yangyin Chushi Decoction may regulate the immune system by regulating inflammatory factors and their associated intestinal flora,exerting anti-inflammatory and osteoprotective effects.关键词
