越婢汤对阿霉素诱导的人脐静脉内皮细胞高通透性的影响及机制研究
Study on the Effects and Mechanism of Yuebi Decoction on Adriamycin-induced Hyperpermeability of Human Umbilical Vein Endothelial Cells

Abstract
Objective To investigate the effects of Yuebi Decoction on adriamycin-induced hyperpermeability of human umbilical vein vascular endothelial cells(HUVEC);To explore the possible mechanism of its therapeutic effect on edema in nephrotic syndrome.Methods HUVEC were divided into control group,model group and Yuebi Decoction low-,medium-,and high-dosage groups(200,300,400 μg/mL).CCK-8 method was used to detect cell viability,Transwell method was used to detect cell permeability,Western blot was used to detect the expression of apoptosis signal regulated kinase 1(ASK1),zonula occludens-1(ZO-1),vascular endothelial-cadherin(VE-cadherin),and p38 protein,immunofluorescence method was used to detect the expression of ZO-1 and VE-cadherin.Results Compared with the control group,the cell activity of HUVEC in the model group was significantly decreased,cell permeability significantly increased,and cell swelling and membrane structure damage occurred,the expressions of ASK1 and p-p38 protein significantly increased,while the expressions of ZO-1 and VE-cadherin protein were significantly decreased(P<0.01).Compared with the model group,the cell activity and permeability of Yuebi Decoction low-,medium-,and high-dosage groups significantly increased,and the expressions of ASK1 and p-p38 protein were significantly decreased,and the expressions of ZO-1 and VE-cadherin protein significantly increased(P<0.05,P<0.01).Conclusion Yuebi Decoction can improve the high permeability of HUVEC induced by adriamycin,and its mechanism may be related to the inhibition of ASK1/p38 signaling pathway and upregulation of cell junction protein expression.关键词
