Optimization of Extraction Process and Antioxidant Activity in Vitro of Aralia elata Polysaccharides

Abstract
Objective The aqueous extraction process of Aralia elata polysaccharides was optimized and its antioxidant activity in vitro was preliminarily investigated.Method The effects of material-liquid ratio,extraction temperature,extraction time and number of extractions on the yield of Aralia elata polysaccharides were optimized by using a one-way response surface methodology;The composition of various components in Aralia elata polysaccharides was detected by using phenol sulfuric acid method,meta hydroxybiphenyl method and Coomassie brilliant blue method;The antioxidant activity of Aralia elata polysaccharides was investigated by the scavenging ability of free radicals in vitro,and the antioxidant effects of Aralia elata polysaccharides were investigated by constructing PC12 cellular oxidative damage model.Results The polysaccharide yield was predicted to be 4.38%and the actual polysaccharide yield was up to 4.29% with material-liquid ratio m(g)∶ V(mL)=1∶10 and 3 times at 91℃ for 3.2 h/time.The content of the polysaccharide obtained from the response surface optimization was 21.39%,the protein content was 7.9%,and the content of glucuronide was 1.08%;Aralia elata polysaccharide can effectively remove DPPH,superoxide anion and hydroxyl radicals,and it can significantly increase the intracellular SOD activity (P<0.05,P<0.01),reduce the MDA content (P<0.05),and increase the survival rate of the cells in oxidatively damaged PC12 cells.Conclusion The extraction method is stable and reliable,and the prepared Aralia elata polysaccharide has obvious antioxidant activity in vitro,which can provide theoretical and experimental basis for the prevention and treatment of AD.关键词
