Abstract
This study focuses on autophagy and liver lipid metabolism in chickens. Firstly, three groups of different concentrations (2.5 mmol/L, 5 mmol/L and 10 mmol/L) were set to treat chicken hepatoma cell lines (LMH) at different concentrations (2.5 mmol/L, 5 mmol/L and 10 mmol/L) for 24 h and 48 h, respectively. The total RNA of the cells was extracted, and the expression of autophagy-related genes at the mRNA level was detected by RT-qPCR to screen the optimal inhibitory concentration and time of 3-MA. Then, the expression of genes related to lipid metabolism in chicken LMH cells treated with optimal conditions 3-MA was detected. The results show that after 24 h treatment of chicken LMH cells with 3-MA at 2.5 mmol/L, the expression of genes Atg3, Map1lc3a and Becn1 was down-regulated at the mRNA level, with the optimal condition being consistent with expectations. Compared with the control group, the expression of genes related to lipid metabolism was significantly down-regulated, the expression level of FASN was significantly up-regulated, and the expression level of Fabp7 and FOXO1 was significantly changed compared with the control group. Therefore, a certain concentration of the autophagy inhibitor 3-MA can promote lipid metabolism in vitro.关键词
