摘要

Abstract

Objective To study the antiviral efficacy of baicalin on SARS-COV-2 entry process in vitro.Methods The SARS-CoV-2 S pseudo-typed virions containing a luciferase reporter gene and Luciferase kit were used to detect the changes of fluorescein expression caused by baicalin in Huh-7 cells after incubation with the pseudo-typed virion,and then the virus inhibition curve is plotted.Results Baicalin can significantly inhibited the infection rate of pseudo virus.There was no signifi-cant difference in the virus inhibition curve between the baicalin&virus pre-incubation group and co-incubation at different concentrations,indicating that baicalin could not directly bind to virus,but could play an antiviral role by affecting the cell fu-sion process mediated by the virus S protein.We further found that,the inhibition rate of baicalin to virus decreased signifi-cantly in the 4 h group,but had no significant difference in the 0 h and 2 h groups at the concentration of 0.125 mg·mL-1,in-dicating that baicalin may have an inhibitory effect on virus invasion stage rather than adsorption stage,and the mediated inhi-bition stage occurred within 4 h.Conclusion Baicalin may mediate the fusion of SARS-CoV-2 S protein with cell surface re-ceptor and exert anti-SARS-CoV-2 activity by playing a role in inhibiting virus invasion in non-adsorption stage.
