Development of a New Method for Dyeing Polystyrene Microplastics with Nile Red

对微塑料的分析研究需从量化方法开始.目前,针对微塑料常使用的染色方法都存在荧光强度较低和染色分布不均匀的缺点.文章采用了旋转蒸发法(旋蒸法)和尼罗红(NR)染料对聚苯乙烯微塑料(PS)进行染色.采用扫描电镜(SEM)和红外光谱法(FTIR)对比染色前后PS的微观结构和表面基团的变化,利用流式细胞仪对荧光PS(FPS)的数量进行统计并与荧光法测定结果对照.结果表明:旋蒸法对PS的染色效果符合预期,定量分析小尺寸FPS的相对标准偏差在10%以内,且FPS的荧光信号强,易于检测.在FPS的稳定性测试中发现,该方法的染色效果较稳定,适用于对不同尺寸的PS和在模拟环境中的样品进行染色.

The research of microplastics should start with quantitative analysis methods.At present,the commonly used dyeing methods for microplastics have the disadvantages of low fluorescence intensity and uneven dyeing distribution.Polystyrene microplastics(PS)were dyed by rotary evaporation and Nile Red(NR)dyes in this paper.Scanning electron microscope(SEM)and infrared spectroscopy(FTIR)were used to compare the changes of the microstructure and surface groups of PS before and after staining.The quantity of fluorescent PS(FPS)was counted by flow cytometry and compared with the results measured by fluorescence method.The results show that the dyeing eccifency of rotary evaporation on PS is in line with expectations,the relative standard deviation of quantitative analysis of small size FPS is less than 10%,and the fluorescence signal of FPS is strong and easy to detect.In the stability test of FPS,it is found that the dyeing effect of this method is stable,and it is suitable for dyeing PS of different sizes and samples in simulated environment.the dyeing efficiency is stable,and this method is suitable for dyeing PS of different sized and samples in simulated environment.
