摘要

Abstract

Objective:To investigate the effect of exogenous basic fibroblast growth fac-tor(bFGF)on contractility of human uterine smooth muscle cells(HUt SMCs)and its possible mechanism.Methods:Exogenous bFGF of different concentrations were used to treat HUt SMCs.The mRNA levels of connexin 43(CX43)and oxytocin receptor(OXTR)were detected by qRT-PCR,and the expressions of CX43 and OXTR proteins were detected by Western blot.Collagen contractile test was used to study the effect of exogenous bFGF on the contractility of HUt SMCs.The level of intracellular Ca2+ was observed by laser confocal microscope.HUt SMCs were treated with exogenous bFGF and JSH-23(NF-κB transcription activity inhibitor),and the expression of CX43 was detected.ELISA was used to detect the levels of IL-6 and IL-8 secreted by HUt SMCs.Results:bFGF could inhibit the expression of contraction-related molecules(CX43,OXTR)mRNA and protein in HUt SMCs.bFGF can inhibit the contractility of HUt SMCs and decrease the concentration of intracellular Ca2+.bFGF upregulates the expression of key proteins of NF-κB pathway and downregulates the expression of CX43 and NF-κB p65 pro-tein.This effect is reversed by JSH-23.Conclusion:bFGF plays a pro-inflammatory role by acti-vating NF-κB pathway and regulating the expression of CX43,which involves in regulating the contractility of HUt SMCs.
