摘要

Abstract

Objective To explore the prediction of tumor immune microenvironment in diffuse large B-cell lymphoma(DLBCL)based on glycolysis-related genes,and to study its potential mechanism.Methods The RNA-seq data and corresponding clinical information of DLBCL patients were downloaded from the NCICCR database.Based on the genes related to glycolysis,they were divided into C1 cluster and C2 cluster by consensus clustering method.Kaplan-Meier(K-M)survival analysis,ESTIMATE,TIMER,and ssGSEA analysis were used to determine differences in survival,function,and tumor microenvironment(TME)between the two identified cluster classes.The potential mechanism was elucidated by GO,KEGG and GSEA functional analysis.Results The survival rates between the two clusters were significantly different.The K-M survival curve showed that the survival rate of C1 cluster was higher than that of C2 cluster,and the immune score,ESTIMATE score and matrix score were higher than those of C2 cluster.GO and KEGG analysis showed that the differentially expressed genes between the two clusters were mainly enriched in extracellular matrix and immune pathways.GSEA analysis showed that the enrichment of immune pathways in C1 cluster was higher than that in C2 cluster.Conclusion Differentially expressed genes in C1 cluster are significantly associated with tumor immune microenvironment,and the survival rate is higher than that in C2 cluster.It indicates that the disorder of glycolysis metabolism leads to the damage of tumor immune microenvironment,which leads to the poor prognosis of DLBCL.
