抑制宿主脂肪酸合成关键酶活性影响O型口蹄疫病毒的复制
Effect on FMDV type O replication by inhibition of host fatty acid synthesis key enzymes activities

Abstract
In order to investigate the effect of fatty-acid biosynthesis in host cells on the replication of foot-and-mouth disease virus type O,the PK-15 cells and the IBRS-2 cells as cell models,respectively,were used to explore the regulation of FMDV replication.The toxic effects of inhibitors on cells were detected by CCK-8 kit to screen the appropriate pretreatment concentration of the in-hibitors.Western-blot and real-time fluorescence quantification(qPCR)were used to detect the regula-tory effects of exogenous addition of C75 and TOFA on FMDV replication.The results showed that,com-pared with the control group,the protein level and transcript level of FMDV were significantly down-regulated after the adding TOFA or C75.In conclusion,inhibition of fatty acid synthesis key en-zymes activities in host cells can reduce efficiency of FMDV replication,which broke a new path for further research on the pathogenic mechanism of FMDV.关键词
