为研究陕西榆林地区牛、羊源大肠杆菌的流行特点及其对氨基糖苷类抗生素的耐药情况,采集746份牛、羊粪便样本进行细菌分离纯化和大肠杆菌特异性引物phoA PCR鉴定,并对鉴定得到的大肠杆菌进行氨基糖苷类抗生素耐药性试验,对5种钝化酶基因AadA1、AbdB、Aph(3′)-Ⅰ、Aac(6′)-Ⅰb、Aac(3′)-Ⅱ进行PCR检测和同源性序列分析.结果显示,榆林地区大肠杆菌总检出率为42.49%;牛、羊源大肠杆菌对6种氨基糖苷类药物的耐药率从高到低依次为链霉素(70.7%)、卡那霉素(64.3%)、阿米卡星(47.9%)、庆大霉素(45.1%)、新霉素(39.4%)和妥布霉素(30.3%);多重耐药率(耐3种及3种以上药物)为67.0%;耐药基因AadA1和Aph(3′)-Ⅰ的检出率分别为79.5%和83.0%.综上表明,榆林地区检出的牛、羊大肠杆菌对氨基糖苷类抗生素的耐药性比较强,耐药基因以AadA1、Aph(3′)-Ⅰ为主.

In order to study the epidemic characteristics of Escherichia coli from cattle and sheep in Yulin,Shaanxi province,and its resistance to aminoglycoside antibiotics,746 fecal samples from cattle and sheep were collected for bacterial isolation and purification,as well as identification by Escherichia coli specific primer phoA PCR.The identified Escherichia coli were tested for aminoglycoside antibiotic resistance,and five passivating enzyme genes AadA1,AbdB,Aph(3′)-Ⅰ,Aac(6′)-Ⅰb,Aac(3′)-Ⅱ were detected by PCR for homology sequence analysis.The results showed that the total detection rate of Escherichia coli in Yulin area was 42.49%.The drug resistance rates of Escherichia coli from cattle and sheep to six aminoglycoside were 70.7%for streptomycin,64.3%for kanamycin,47.9%for amikacin,45.1%for gentamicin,39.4%for neomycin,and 30.3%for tobramycin,respectively.The multi-drug resistance rate(resistance to 3 or more drugs)was 67.0%.The detection rates of drug resistance genes AadA1 and Aph(3′)-Ⅰ were 79.5%and 83.0%respectively.In summary,the resistance of cattle and sheep Escherichia coli to aminoglycoside antibiotics was relatively strong,and the drug resistance genes were mainly AadA1 and Aph(3′)-Ⅰ.
