核纤层蛋白基因在胶质母细胞瘤恶性生物学行为中的作用OACSTPCD
Role of lamin A/C gene in the malignant biological behavior of glioblastoma
目的:研究核纤层蛋白基因(LMNA)在人类胶质母细胞瘤中的表达特点及该基因在胶质母细胞瘤发病过程中的作用机制.方法:分析公共数据库TCGA中LMNA基因在胶质母细胞瘤中的表达及其与患者预后的关系.构建LMNA基因的敲减慢病毒,构建胶质母细胞瘤细胞系U87的LMNA基因稳定敲减株;进一步对U87的LMNA基因敲减株及其对照细胞株进行克隆形成实验和细胞3D成球实验,检测LMNA基因对细胞增殖与成球能力的影响;通过替莫唑胺加药实验检测LMNA基因敲减细胞株对替莫唑胺的耐药性;最后经裸鼠颅内种瘤实验检测LMNA基因敲减在体内对胶质母细胞瘤生长的抑制作用.结果:TCGA数据显示,LMNA基因在胶质母细胞瘤组织中的表达程度显著高于正常组织,且与患者生存预后呈负相关,与肿瘤细胞恶性程度呈正相关.LMNA基因敲减显著降低了U87细胞株的克隆生成及细胞成球能力,显著增加了U87细胞株对替莫唑胺的敏感性;裸鼠颅内种瘤实验显示,LMNA基因敲减的U87细胞株在裸鼠脑内生成的肿瘤体积显著减小,且小鼠存活时间延长.结论:LMNA基因在胶质母细胞瘤中表达增高,与患者生存呈负相关,LMNA基因敲减可抑制U87胶质母细胞瘤细胞株的增殖,并增加对替莫唑胺的敏感性,是胶质母细胞瘤治疗的潜在靶点.
Objective:To study the expression characteristics of lamin A/C(LMNA)gene in human glioblastoma and the mechanisms of its roles in the pathogenesis of glioblastoma.Methods:The study analyzed the expression of LMNA gene in glioblastoma and its correlation with patient prognosis in the public databases of TCGA.The LMNA gene sh-RNA virus was constructed to generate stable LMNA knockdown strains of glioma cell line U87.Then the clone formation and 3D spheroidization experiments were conducted on the LMNA knockdown strains of U87 and its control group to detect the impact of LMNA gene on cell proliferation and spheroidization ability.The resistance of LMNA knockdown U87 to temozolomide was detected through temozolomide treatment assay.The inhibitory effect of LMNA knockdown on glioblastoma growth and proliferation in vivo was detected through intracranial tumor implantation experiments in nude mice.Results:TCGA data showed that the expression level of LMNA in glioblastoma tissue was significantly higher than that in normal tissue,and it was negatively correlated with patient survival prognosis.The expression level of LMNA was positively correlated with the malignancy of tumor cells.LMNA knockdown significantly reduced the clone generation and spheroidization ability of U87 cell line,while it significantly increased the sensitivity of U87 cell line to temozolomide.The intracranial tumor implantation experiments in nude mice showed that the U87 cell line with LMNA knockdown significantly reduced the tumor volume in the mouse brain and improved their survival time.Conclusions:LMNA gene expression is increased in glioblastoma and negatively correlated with patient survival.LMNA gene knockdown can inhibit the proliferation of U87 glioma cell line and increase its sensitivity to temozolomide,making it as a potential target for glioblastoma treatment.
