七氟醚麻醉过程中mPFC脑区不同类型神经元胞外腺苷递质动态变化研究
Change in extracellular adenosine in different types of neurons in the mPFC during sevoflurane anesthesia

Abstract
Objective To investigate the dynamic change of extracellular adenosine release from glutamatergic pyramidal(Pyr)neurons and two subtypes of gamma-aminobutyric acid(GABA)neurons:parvalbumin(PV)neurons and somatostatin(SST)neurons,in the medial prefrontal cortex(mPFC)during sevoflurane anesthesia-induced loss of consciousness and recovery.Methods AAV-CaMk Ⅱ α-Cre and AAV-DIO-Ado1.0 mixed viruses were injected into C57 mice(n=6),AAV-DIO-Ado1.0 virus were injected in SST-IRES-Cre mice(n=6)and PV-IRES-Cre mice(n=6),and were left for 3 weeks for virus transfection.A multichannel fiber photometry system was used to detect and record the fluorescence signals of GRABAdo1.0.Changes in adenosine were re-corded during the loss and recovery of righting reflex in mice during 2.3%sevoflurane anesthesia.Results Sevoflurane was able to cause an increase in extracellular adenosine in pyramidal neurons,SST interneurons,and PV interneurons in the mPFC,which did not immediately decrease with the cessation of sevoflurane inhalation.Conclusion Altered consciousness in sevoflurane anesthesia may be modulated by extracellular adenosine trans-mission in mPFC.关键词
