Abstract

Objective:To optimize the fermentation process and improve its production efficiency of Armillaria mellea M6 strain.Methods:With Armillaria mellea M6 strain as the test strain,the optimum liquid fermentation medium of the strain was screened by sha-king bottle liquid culture.Based on the results of shaker culture,the highly active seed liquid was prepared by multistage homogeniza-tion,and the liquid deep fermentation was carried out to optimize the fermentation conditions.Results:The optimal liquid deep fermenta-tion conditions for Armillaria mellea M6 strain were as follows:rotational speed 150 r/min,inoculation volume 5%,dissolved oxygen 20%and temperature 28 ℃.Under these conditions,the biomass dry weight of Armillaria mellea M6 strain was 18.39 g/L after fermen-tation for 180 hours using liquid fermentation medium formula 1.Using liquid fermentation medium 2 for 120 hours,the biomass dry weight of Armillaria mellea M6 strain was 23.83 g/L.Conclusion:After the optimization of liquid culture conditions,the biomass pro-duction of Armillaria mellea M6 strain is higher.This study promotes the rapid production of Armillaria mellea M6 strainand provides a technical reference for its application in Gastrodia elata cultivation.
