新疆许多规模化牛场存在牛病毒性腹泻病毒(BVDV)的感染,且因病毒基因型具有多样性,给该病的防控、疫苗选择、净化带来了一定的困难,开展新疆地区规模化牛场BVDV基因分型鉴定,对BVDV防控具有重要的指导作用.采集BVDV胶体金抗原检测卡确定为阳性的犊牛抗凝血样品并分离淋巴细胞,在MDBK单层细胞上进行病毒分离,盲传5代,提取培养物RNA,以BVDV5'-UTR、Npro特异性引物进行逆转录-聚合酶链反应(RT-PCR)检测分析.结果显示,获得BL分离株1株,属于BVDV-1m亚型,研究结果丰富了新疆南疆地区规模牛场BVDV分子流行病学数据库.

Many large-scale cattle farms in Xinjiang are infected with bovine viral diarrhea virus(BVDV),and the genotypes are diverse,which brings some difficulties to the prevention and control of the disease,vaccine selection and purification.The genotyping of large-scale cattle farms in Xinjiang plays an important guiding role in the prevention and control of BVDV.In this test,calf anticoagulant samples,confirmed posi-tive for BVDV colloidal gold antigen detection card,were collected and lymphocytes were isolated.Virus i-solation was performed on MDBK monolayer cells,blind transmission for five generations,and trophic RNA was extracted for reverse transcript polymerase chain reaction(RT-PCR)analysis with BVDV 5'-UTR and Npro-specific primers.The results showed that one BL isolate,belonging to the BVDV-lm sub-type,and this study enriched the molecular epidemiology database of BVDV in large-scale cattle farms in southern Xinjiang.
