旨在探究鸭肠炎病毒(DEV)和A型产气荚膜梭菌(CpA)共感染绿壳蛋鸭114 h时十二指肠的差异代谢物变化.试验选取12只健康绿壳蛋鸭,随机分为对照组(CK)和共感染组(TCD).通过临床观察、剖检病理观察以及PCR检测确定DEV和CpA共感染成功,再使用液相色谱-质谱联用(LC-MS)检测CK组和TCD组感染114 h时鸭十二指肠的代谢物变化,筛选潜在差异代谢物及代谢通路.结果表明,与CK组相比,TCD组鸭十二指肠感染114 h时的总差异代谢物29种,主要有5'-s-甲基-5'-硫腺苷、胞苷、脱氧皮质酮21-葡萄糖苷、腐胺和2-氧吲哚、腐胺、柚皮素、2-氧吲哚、吲哚-3-乙醛等;差异代谢物通过KEGG通路富集分析,共显著富集到22条代谢通路,主要有烟酸酯和烟酰胺代谢、半胱氨酸和蛋氨酸的代谢、酪氨酸代谢、色氨酸代谢、甘油磷脂代谢等,其中与机体免疫和炎症相关的有腐胺、柚皮素、2-氧吲哚、吲哚-3-乙醛、5'-s-甲基-5'-硫腺苷、烟酸脂和半胱氨酸-蛋氨酸代谢、色氨酸代谢和烟酰胺代谢等.综上所述,DEV和CpA共感染绿壳蛋鸭后,可能通过影响腐胺和柚皮素等代谢物以及色氨酸代谢和半胱氨酸-蛋氨酸代谢通路来影响绿壳蛋鸭十二指肠的炎症,为阐明DEV和CpA共感染机制提供科学依据.

The aim of this experiment was to investigate the changes of differential metabolites in the duode-num at 114 h of co-infection of green-shelled egg-laying ducks with duck enteritis virus(DEV)and Clos-tridium perfringens type A(CpA).Twelve healthy green-shelled egg ducks were selected for the experi-ment and randomly divided into the control group(CK)and the co-infected group(TCD).We determined the success of DEV and CpA co-infection through clinical observation,pathologic observation by dissection and PCR,and then examined the metabolite changes in the duodenum of ducks in the CK group and TCD groups at 114 h of infection by liquid chromatography-mass spectrometry(LC-MS),in order to screen for the potential differential metabolites and metabolic pathways.The results showed that 29 different metabo-lites were found in the duodenum of ducks in the TCD group at 114 h of infection compared with those in the CK group,including 5'-s-methyl-5'-thioadenosine,cytidine,deoxycorticosterone 21-glucoside,putrescine and 2-oxoindole,putrescine,naringenin,2-oxoindole,and indole-3-glycolide,etc.The metabolic pathways,mainly nicotinic acid,niacin,niacin,2-oxoindole,and indole-3-glycolide,were significantly enriched into 22 metabolic pathways via KEGG enrichment.Metabolic pathways,mainly nicotinic acid ester and nicotinam-ide metabolism,cysteine and methionine metabolism,tyrosine metabolism,tryptophan metabolism,glycer-ophospholipid metabolism,etc.Among them,those related to the body's immunity and inflammation were putrescine,naringenin,2-oxoindole,indole-3-glycolaldehyde,5'-s-methyl-5'-thioadenosine,nicotinic acid es-ter,cysteine Gmethionine metabolism,tryptophan metabolism and nicotinamide metabolism.In summary,DEV and CpA co-infection in green-shelled ducks may affect duodenal inflammation in green-shelled ducks by influencing metabolites such as putrescine and naringenin,as well as tryptophan metabolism and cyste-ine-methionine metabolism pathways,which provides a scientific basis for elucidating the mechanism of co-infection with DEV and CpA.
