摘要

Abstract

Objetive The aim of the present study was to explore the possible mechanism of 4 weeks treadmill exercise preintervention in improving cognitive function of cerebral ischemia reperfusion(CI/R)rats by measuring the cerebral infarction volume,hippocampus inflammatory biomarkers levels and the synaptic proteins expression in hippocampus CA1.Methods 40 SD male rats were modeled for CI/R and the CI/R exercise group rats were subjected to 4 weeks of incremental load treadmill exercise before modeling.After 72 h of CI/R,the learning and memory ability of rats were evaluated,the cerebral infarction volume%,the level of TNF-α,IL-6 in hip-pocampus and the expression of SYN and PSD-95 in hippocampal CA1 region were measured.Results 1)Compared with the sham operation group,the escape latency significantly increased and the number of crossing the platform significantly decreased in the morris water mazet test;cerebral infarction volume%and the level of IL-6 and TNF-α in hippocampus significantly increased,while the MOD of SYN and PSD-95 in hippocampus CA1 significantly decreased.2)The 4-week treadmill exercise preintervention can signifi-cantly improve the learning and memory ability of CI/R rats,the cerebral infarction volume%and the TNF-α level in hippocampus significantly decreased;the MOD of SYN and PSD-95 had a significant up-regulation in hippocampus CA1 region in CI/R exercise group rats when compared with the CI/R model group.Conclusion Treadmill exercise preintervention might prevent memory deficits after CI/R through reducing hippocampal neuroinflammatory response,reducing cerebral infarction volume and increasing the synaptic associated protein expression in hippocampal CA1,which is considered to be an effective strategy to prevent ischemic injury.
