奶牛妊娠中后期胎盘mRNA差异表达基因分析OA北大核心CSTPCD
Analysis of Differentially Expressed Genes of Placental mRNA in Dairy Cows during Mid and Late Pregnancy
奶牛的繁殖问题是制约我国奶业快速发展的瓶颈.为了研究奶牛妊娠中后期胎盘差异基因表达情况,探索mRNA在奶牛妊娠过程中的调控作用,本研究以奶牛妊娠中后期胎盘、非妊娠对照组子宫为对象,使用RNA-seq测序技术进行转录组分析.结果发现,与非妊娠对照组相比,妊娠中期和后期胎盘分别有4 333和4 354个基因的表达出现显著变化,其中上调的基因分别有2 328和2 519个,下调的基因分别有2 005和1 835个;778个差异基因只存在于妊娠中期,757个差异基因只存在于妊娠后期.GO分析结果显示,妊娠中后期差异表达的基因主要在定位、生物粘附、细胞连接、生殖等过程.KEGG分析结果显示,妊娠中后期差异表达的基因在ECM-受体相互作用、细胞粘附分子、细胞周期、粘着斑、PI3K-Akt信号通路、癌症的途径以及蛋白质的消化吸收等通路.与非妊娠奶牛相比,妊娠中后期胎盘基因存在明显的差异表达,其中PLET1、CSH1、SULT1E1、PRP1、LPAR2、CSF1R、PDCD1等基因表达明显上调,推测其可能参与奶牛的妊娠调控,在一定程度上影响着妊娠的发生.本研究结果为深入挖掘奶牛妊娠中后期的分子调控机制提供了理论基础.
Reproduction of dairy cows is a bottleneck that restricts the rapid development of China's dairy industry.In order to study the differential gene expression in the placenta of cows in mid and late gestation,and to explore the regulatory role of mRNA in the process of cow pregnancy,the placenta of cows in mid and late gestation,and the uterus of non-pregnant control group was used as the study objects,and transcriptome analysis was performed using RNA-seq sequencing technology.The results revealed that 4 333 and 4 354 genes showed significantly differential expressions in the mid and late-gestation placentas,respectively,compared with the non-pregnant control group,with 2 328 and 2 519 genes up-regulated and 2 005 and 1 835 down-regulated genes,respectively.778 differentially expressed genes existed only in the mid-pregnancy,and 757 differentially expressed genes existed only in the late-pregnancy.GO analysis indicated that the differentially expressed genes in mid and late gestation were mainly enriched in the processes of localization,bioadhesion,cell junction,and reproduction.KEGG analysis showed that the differentially expressed genes in mid and late gestation were enriched in the pathways of ECM-receptor interactions,cell adhesion molecules,cell cycle,adhesion patches,PI3K-Akt signaling pathway,pathways of cancer,and digestion and absorption of proteins.Between the middle and late stages of pregnancy,there were obvious differences in the expression of genes,including PLET1,CSH1,SULT1E1,PRP1,LPAR2,CSF1R,PDCD1,which were up-regulated,and it was hypothesized that they might be involved in the regulation of pregnancy in cows,and influence the occurrence of pregnancy to a certain extent.The results of this study provide a theoretical basis for the in-depth excavation of the molecular regulatory mechanisms in the middle and late stages of pregnancy in dairy cows.
