脯氨酸对山羊精液低温保存的影响OA北大核心CSTPCD
The Effect of Proline on Goat Semen Storage at Low Temperature
为提高精液对精子的抗氧化保护能力,延长精子在体外的存活时间,在低温保存(4℃)条件下,向山羊精液基础稀释液中添加浓度为0、0.5、1、2、4 mM的脯氨酸,经保存24 h、48 h后,检测各处理组的精子活力、顶体完整率、质膜完整率,及48 h各组线粒体活性、活性氧水平、丙二醛、脂质过氧化物含量、抗氧化酶活性等精子质量评定指标.结果表明,在精液中添加一定浓度的脯氨酸可提高精子抗氧化能力,且添加最适浓度为1 mM.在1 mM处理组中,精子48 h总活力71.3%,比对照组显著提高18.57%(P<0.01),顶体完整率提高12%,质膜完整率提高10.5%,线粒体活性提高23.15%,抗氧化酶活性均显著提高(P<0.05),活性氧水平、丙二醛、脂质过氧化物含量均显著降低(P<0.05),并且脯氨酸脱氢酶活性显著提高(P<0.05),凋亡相关蛋白(Caspase-3、Caspas-9、P53和BCL-2)表达量显著降低(P<0.05).因此,在4 ℃液态保存的山羊精液中,添加脯氨酸能够提高保存后的精液品质.这为脯氨酸作为一种可以有效提高山羊精液品质的抗氧化剂提供了新的思路.
To improve the antioxidant protection capacity of sperm and extend the viability of sperm in vitro,concentrations of 0,0.5,1,2,and 4 mM of proline were added to the goat semen basic dilution medium under low-temperature storage conditions(4 ℃).After 24 and 48 hours,the sperm motility,acrosome integrity,and plasma membrane integrity were evaluated in each treatment group.Additionally,mitochondrial activity,reactive oxygen species level,malondialdehyde,lipid peroxide content,antioxidant enzyme activity and other sperm quality assessment indexes were also investigated after 48 h of preservation.The results showed that addition of proline to the diluted medium improved the sperm antioxidant capacity,and the optimal concentration was 1 mM.In the 1 mM treatment group,the sperm total motility at 48 h was 71.3%,significantly increased by 18.57%compared with the control group(P<0.01),meanwhile,acrosome integrity was increased by 12%,plasma membrane in-tegrity was increased by 10.5%,the mitochondrial activity was increased by 23.15%,and antioxidant en-zyme activity were also significantly increased(P<0.05),while reduction of sperm ROS level,malondial-dehyde level,lipid peroxide were overserved in proline treatments(P<0.05).Furthermore,addition of proline significantly increased the proline dehydrogenase activity(P<0.05),it also significantly decreased the expression of the apoptosis proteins such as Caspase-3,Caspase-9,P53 and BCL-2(P<0.05).There-fore,addition of proline to the diluted medium of goat semen preserved at 4oC improves the goat sperm quality.These findings provide novel insights that proline can serve as an effective antioxidant to improve the goat sperm quality when preserved at low temperature.
