Effect of ADSCs-loaded GelMA hydrogel on wound healing in diabetic rats

Abstract
Objective To investigate the effect of adipose-derived stem cells(ADSCs)-loaded Gelatin Methacrylate(GelMA)hydrogel scaffolds on skin wound healing in diabetic rats.Methods ADSCs were seeded on the surface of GelMA scaf-folds.Streptozocin was used to establish a rat model of diabetes,and a round full-thickness skin defect with a diameter of 2 cm was created on the back of the rats.The rats were divided into control group(without any treatment),GelMA group(treated with scaf-folds alone),and ADSCs-loaded GelMA group(treated with ADSCs-loaded scaffolds).Wound healing was observed for each group;HE staining was used to observe the histology of skin tissue;immunohistochemical staining was used to measure the ex-pression of transforming growth factor-beta 1(TGF-β1).Results Compared with the control group and the GelMA group,the ADSCs-loaded GelMA group had a higher wound healing rate at 14 and 21 days after surgery,a stronger expression of TGF-β1 at 14 days after surgery,and a reduction in capillary bleeding at 21 days after surgery.Conclusion ADSCs-loaded GelMA hydrogel scaffolds can promote wound healing in diabetic rats,and TGF-β1 may play an important role in this process.关键词
