NLRP3炎症小体在人骨关节炎软骨中的表达及作用
Expression and role of NOD-like receptor family pyrin domain containing 3 inflammasome in human osteoarthritis cartilage

Abstract
Objective To investigate the expression and role of NOD-like receptor family pyrin domain containing 3(NLRP3)inflammasome in human osteoarthritis(OA)cartilage.Methods A total of 30 cartilage samples were collected from the patients with OA,and 10 normal cartilage samples were collected.RT-qPCR was used to measure the mRNA expression levels of the NLRP3 inflammasome components NLRP3,apoptosis-associated speck-like protein containing CARD(ASC),and cysteinyl aspartate specific proteinase-1(caspase-1)and the inflammatory factor interleukin-1β(IL-1β);Western blot was used to measure the protein expression levels of NLRP3,ASC,and caspase-1;ELISA was used to measure theprotein expression level of IL-1β.Results Compared with the normal cartilage group,the OA group had significant increases in the mRNA and protein expression levels of NLRP3,ASC,and caspase-1 in cartilage tissue(t=4.042-6.704,P＜0.001).Compared with the normal cartilage group,the OA group had significant increases in the mRNA and protein expression levels of IL-1β(t=4.826,6.144,P＜0.001).Conclusion There are increases in the expression of NLRP3 inflammasome components and IL-1β in OA,which are involved in the formation of inflammatory environment in OA articular cartilage.关键词
