摘要

Abstract

Objective:To observe the effects of silent information regulator 2 family member 5(SIRT5)gene on inva-sion,metastasis and glycolysis of ovarian cancer cells,and explore its potential molecular mechanisms.Methods:Reverse transcription polymerase chain reaction and Western blot were used to analysis SIRT5 expressions in various ovarian cancer cell lines and normal ovarian epithelial cells.Small interfering RNA(siRNA)technology was used to perform the knockdown of SIRT5 gene.Transwell and wound healing assay were used to detect the influence of SIRT5 gene knockdown on the invasion and migration of ovarian cancer cells.Western blot was used to detect the expressions of key proteins in epithelial mesenchymal transition(EMT),glycolytic pathway and STAT3/c-MYC signaling pathway.Results:The levels of SIRT5 mRNA and protein in all ovarian cancer cell lines were higher than those in normal ovarian epithelial cells(P<0.05);siRNA technology could significantly reduce both the transcriptional and protein expression levels of SIRT5(P<0.05).SIRT5 knockdown could significantly inhibit the migration and invasion ability(P<0.05)and the glycolysis level(P<0.01)of ovarian cancer cells,promote the expression of EMT-related protein E-cadherin,inhibit the expressions of N-cadherin and Vimentin,reduce the expressions of glycolysis-related proteins PKM2,GLUT1 and LDHA,and down-regulate STAT3 phosphorylation and c-MYC protein levels(P<0.05).Conclusion:Knockdown of SIRT5 gene can inhibit the migra-tion and invasion ability and glycolysis level of ovarian cancer cells,negatively regulate the EMT process and glycolysis path-way,which may be related to the inhibition of STAT3/c-Myc signaling activation.
