基于生信分析探索IL4I1表达对卵巢癌的影响OACSTPCD
Effectof IL4I1 Expression on Ovarian Cancer Based on Bioinformatics Analysis
目的:探讨白细胞介素4诱导蛋白1(interleukin-4 induced 1 protein,IL4I1)在卵巢癌中的表达情况及临床意义.方法:基于TCGA、GTEx数据库,分析IL4I1基因在卵巢癌组织与正常卵巢组织中的表达差异,从HPA数据库获得免疫组化图像,采用RNA-seq检测IL4I1在卵巢癌中的表达水平.使用Kaplan-Meier plotter绘制IL4I1的生存分析图以解读其对卵巢癌患者预后的作用.使用GSEA探索IL4I1相关的生物学功能和信号通路.利用TIMER数据库和CIBERSORT算法分析IL4I1的表达与免疫细胞浸润水平的相关性.结果:IL4I1在卵巢癌组织中表达增加(P<0.001),并与卵巢癌患者较差的无进展生存期相关(P<0.05);GSEA结果提示,IL4I可能参与调节IL6-JAK-STAT3、PI3K-AKT-MTOR通路.TIMER分析结果提示,IL4I1表达水平与B细胞(r=0.190,P<0.05)、CD4+T细胞(r=0.289,P<0.05)、中性粒细胞(r=0.373,P<0.05)、树突状细胞(r=0.322,P<0.05)浸润水平成正相关.结论:IL4I1在卵巢癌中高表达,它可能通过PI3K-AKT-MTOR、IL6-JAK-STAT3通路促进卵巢癌的发生和发展,并与卵巢癌肿瘤免疫具有一定关联,具有作为卵巢癌诊断和治疗靶点的潜力.
Objective:The purpose of this study was to examine how interleukin-4 induced-1 protein(IL4I1)is expressed in ovarian cancer(OC)and its clinical implication.Methods:Relevant data were obtained from TCGA and GTEx databases to analyze the differential expression of IL4I1 in OC tissue and normal ovarian tissue.Immunohistochemical images were taken from the HPA database,and RNA-seq was used to detect the expression level of IL4I1 in OC.And its impact on the prognosis of OC patients was assessed by Kaplan-Meier plotter.Gene set enrichment analysis(GSEA)was used to explore IL4I1-related biological functions and signaling pathways.Correlation between IL4I1 expression and the level of immune cell infiltration was analyzed based on the CIBERSORT algorithm using the TIMER database.Results:IL4I1 expression in OC tissue was significantly higher than that in normal ovarian tissue(P<0.001),and IL4I1 was linked to poor progression-free survival(PFS)in OC patients(P<0.05).GSEA results suggested that IL4I1 may be involved in regulating IL6-JAK-STAT3 and PI3K-AKT-MTOR pathways.TIMER results revealed that IL4I1 expression level was positively correlated to the infiltration levels of B cells(r=0.190,P<0.05),CD4+T cells(r=0.289,P<0.05),neutrophils(r=0.373,P<0.05),and dendritic cells(r=0.322,P<0.05)in OC.Conclusion:IL4I1 is highly expressed in OC and is related to the tumor immunity of OC,it may promote the occurrence and development of OC through IL6-JAK-STAT3 and PI3K-AKT-MTOR pathways,which has potential as a diagnostic and therapeutic target for OC.
