Expression and mechanism of membrane-associated ring finger protein 7 in epithelial ovarian cancer tissues and cells

Abstract
Objective:To investigate the expression and mechanism of membrane-associated ring finger protein 7(MARCH7)in epithelial ovarian cancer(EOC)tissues and cells.Methods:A total of 32 patients who underwent sur-gical treatment were enrolled in this study,including 16 patients with EOC and 16 patients with normal ovarian tis-sues.Ovarian tissues from EOC patients and 16 patients with normal ovarian tissues were collected,and the expres-sion of MARCH7 mRNA was detected by RT-qPCR.After ovarian cancer cells(SKOV3,CAOV3)with overexpres-sion or knockdown of MARCH7 were constructed,Trans well assay was used to observe the effect of MARCH7 on the migration and invasion of EOC cells,and Western blot was used to detect EMT-related proteins E-cadherin,N-cadherin,Snail1 and Slug in MARCH7 overexpressed/down-regulated EOC cells.Results:The expression of MARCH7 mRNA in ovarian cancer tissues was significantly higher than that in normal ovarian tissues(P<0.05).In SKOV3 and CAOV3 cells,there was no significant difference in the migration and invasion ability between the normal control group and the low expression control group and the overexpression control group,as well as the ex-pression of MARCH7 and EMT related proteins E-cadherin,N-cadherin,Snail1 and Slug(all P>0.05);compared with the low expression control group,the migration and invasion abilities of MARCH7 knockdown cells were de-creased,the expression of E-cadherin protein was increased,and the expressions of N-cadherin,Snail1 and Slug pro-tein were decreased(all P<0.01);compared with the overexpression control group,the migration and invasion abili-ties of MARCH7 overexpression cells were increased,the expression of E-cadherin protein was decreased,and the ex-pressions of N-cadherin,Snail1 and Slug protein were increased(all P<0.01).Conclusion:MARCH7 is highly ex-pressed in EOC tissues and cells,which may promote the invasion and migration of EOC cells by promoting the EMT process.关键词
