Identification,Biological Characteristics and Sensitivity to Different Fungicides of the Pathogen Causing Stem Canker of Pinus bungeana
摘要
Abstract
In recent years, stem canker has occurred in a large area in Pinus bungeana cultivation regions of Shaanxi Province, and it seriously affects the economic and ecological value of P.bungeana.To identify the pathogen causing stem canker of P.bungeana, tissue separation method and Koch's postulate were applied to obtain the pathogen of P.bungeana stem canker. It was then identified as Nothophoma quercina by his-tologic sections, morphological characteristics and analyses of ITS,RPB2 and β-tubulin sequences.N.quercina showed the fasted growth rate on the carrot solid medium.Starch was the optimum carbon source, while peptone and beef extract were the optimum nitrogen sources. The optimum temperature and pH for the growth of N.quercina was 25 ℃ and 5-8.The in vitro toxicity tests of fungicides showed that N.quercina exhibited different sensitivities to all the nine tested fungicides.N.quercina showed higher sensitivity to all the tested chemical fungicides except for metalaxyl.N.quercina exhibited the highest sen-sitivity to difenoconazole with a lowest EC50 value of 0.108 mg·L-1,followed by hexaconazole,azoxys-trobin and carbendazim with their corresponding EC50 values of 0.212,0.625 and 0.628 mg·L-1,respec-tively.Among biogenic fungicides,N.quercina showed the highest sensitivity to berberine and osthole with their corresponding EC50 values of 11.914 and 17.375 mg·L-1.The present study can provide guidance for the effective control of P.bungeana stem canker.关键词
