秦岭杜鹃扦插生根及相关生理生化分析研究
Rooting and Related Physiological and Biochemical Analysis of Rhododendron tsinlingense

Abstract
In order to explore the method suitable for cutting and rooting of Rhododendron tsinlingense,this experiment used different cutting methods and different concentrations,types and soaking time of plant growth regulators to explore their effects on the rooting of cuttings.By analyzing the changes of rooting rate,rooting number,longest root length,soluble sugar content,soluble protein content,total nitrogen con-tent,polyphenol oxidase(PPO)activity,indole-acetic acid oxidase(IAAO)activity and peroxidase(POD)activity,the optimal scheme was selected.The results showed that the Nearing method was the most suit-able for the cutting rooting of R.tsinlingense,and the rooting rate was 66%.In orthogonal test,the best cutting effect was obtained by soaking 100 mg·L-1 IBA for 3 h,and the rooting rate was 66%.The physi-ological and biochemical indexes of cuttings were measured.Among different cutting methods,the contents of soluble sugar,soluble protein and total nitrogen of Nearing were the highest,which were 44.151 μg· g-1,2.403 mg·g-1 and 6.267 g·(100 g)-1,respectively.POD and PPO activities of Nearing type were the highest,42.706 U·g-1·min-1 and 1.10 U·mL-1,respectively.The activity of IAAO was the high-est,682.33 μg·(g·h)-1.In the orthogonal test,the highest soluble sugar content was 97.707 µg·g-1 when the cutting was soaked with 400 mg·L-1 NAA and 400 mg·L-1 IBA for 0 h.When soaked in 100 mg·L-1 IBA for 3 hours,the soluble protein,total nitrogen content,and POD and PPO activities reached their highest values,which were 2.313 mg·g-1,436.67 g·(100 g)-1,42.567 U·(g·min)-1,and 1.10 U·mL-1,respectively.The highest IAAO activity was 651 μg·(g·h)-1 when 200 mg·L-1 NAA was soaked for 6 h.Therefore,in different cutting methods,Nearing cutting is easier to take root and the physi-ological indexes reach the maximum,and the effect of cuttings is better when the cuttings are soaked in 100 mg·L-1 IBA for 3h than that of NAA.关键词
