目的:探讨人卵丘颗粒细胞(CGCs)线粒体与胚胎质量及发育潜能的关系.方法:选取2022 年10 月到2023 年10 月于联勤保障部队第九四〇医院生殖中心行IVF/ICSI助孕的60 例患者,收集患者取卵当日CGCs并体外培养,根据取卵后第3d(D3)CGCs对应的优质胚胎率将其分为3 组:A组(优质胚胎率<1/3)、B组(1/3<优质胚胎率>2/3)、C组(优质胚胎率>2/3).通过观察并比较各组间 CGCs 线粒体分布、线粒体膜电位(MMP)、活性氧(ROS)水平、线粒体DNA(mtDNA)拷贝数与细胞内三磷酸腺苷(ATP)水平的差异,评估它们对胚胎质量的影响.结果:与C组相比,A、B组CGCs ATP、MMP明显降低、ROS水平明显升高,差异均有统计学意义(P<0.05).三组间线粒体分布率及mtD-NA拷贝数差异无明显统计学意义(P>0.05).结论:CGCs的ATP、MMP、ROS水平可用于预测胚胎质量.

Objective:To explore the relationship between human cumulus granulosa cells(CGCs)mitochondria and embryo quality and developmental potential.Methods:From October 2022 to October 2023,60 patients who underwent IVF/ICSI assisted pregnancy in the Reproductive Center of the 940th Hospital of the Joint Logistics Support Force were selected as the research objects.The CGCs on the day of egg retrieval were collected and cultured in vitro,and they were divided into three groups according to the high-quality embryo rate corresponding to CGCs on the third day(D3)after egg retrieval:Group A(high quality embryo rate<1/3),Group B(1/3<high quality embryo rate>2/3)and Group C(high quality embryo rate>2/3).By observing and comparing the differences between CGCs in mitochondrial distribution,mito-chondrial membrane potential(MMP),reactive oxygen species(ROS)level,mitochondrial DNA(mtDNA)copy number and intracellular adenosine triphosphate(ATP)level,their effects on embryo quality were evaluated.Results:Compared with group C,the ATP、MMP of CGCS in group A and group B decreased significantly,and the level of ROS increased significantly,with statistical significance(P<0.05).There was no significant difference in mitochondrial distribu-tion rate and mtDNA copy number among the three groups(P>0.05).Conclusion:The levels of mitochondrial ATP,MMP and ROS in human cumulus granulosa cells can be used to predict embryo quality.
