抗磷脂抗体损害子宫蜕膜血管内皮细胞血管形成的机制研究

Abstract
Objective:To study the effect of antiphospholipid antibody(aPL)on the proliferation,migration and angiogenesis of decidual vascular endothelial cells(DVECs).Methods:DVECs were identified by immunofluorescence assay.CCK-8 assay was used to detect the effect of aPL on DVECs proliferation.The effect of aPL on DVECs migration and angiogene-sis was observed by cell scratch assay and angiogenesis assay.Real-time PCR(RT-PCR)was used to detect the effect of aPL on MMP-2 and VEGF mRNA expression in DVECs.Western blot was used to detect the effect of aPL on ERK phosphorylation,VEGF and MMP-2 protein ex-pression in DVECs.Results:aPL inhibited the proliferation,migration and angiogenesis of DVECs.aPL can inhibit ERK phosphorylation,VEGF and MMP-2 expression in DVECs.The expression levels of p-ERK,VEGF and MMP-2 in DVEC cells were significantly increased after the addition of ERK agonists.Conclusion:aPL can reduce the expressions of VEGF and MMP-2 in DVECs by regulating ERK pathway,and then inhibit the proliferation,migration and angio-genesis of DVECs.关键词
