抗磷脂抗体损害子宫蜕膜血管内皮细胞血管形成的机制研究OACSTPCD
Study on mechanism of antiphospholipid antibody damage to the angiogenesis capacity of decidual vascular endothelial cells
目的:研究抗磷脂抗体(aPL)对蜕膜组织血管内皮细胞(DVEC)增殖、迁移与血管形成能力的影响.方法:采用细胞免疫荧光鉴定DVEC细胞;CCK-8 法检测aPL对DVEC细胞增殖的影响;细胞划痕实验与血管形成实验检测aPL对DVEC细胞迁移与血管生成的影响;实时荧光定量PCR(RT-PCR)检测aPL对DVEC细胞基质金属蛋白酶 2(MMP-2)和血管内皮生长因子(VEGF)mRNA 表达的影响;Western blot 检测 aPL 对DVEC细胞中p-ERK、t-ERK、VEGF与MMP-2 蛋白表达的影响.结果:aPL能抑制DVEC细胞增殖、迁移与血管形成能力;aPL能抑制DVEC细胞中p-ERK、VEGF与MMP-2 表达水平,加ERK激动剂后DVEC细胞中p-ERK、VEGF与MMP-2 表达水平明显升高.结论:aPL可通过调控ERK通路减少DVEC细胞中VEGF与MMP-2 表达,进而抑制DVEC的增殖、迁移与血管形成能力.
Objective:To study the effect of antiphospholipid antibody(aPL)on the proliferation,migration and angiogenesis of decidual vascular endothelial cells(DVECs).Methods:DVECs were identified by immunofluorescence assay.CCK-8 assay was used to detect the effect of aPL on DVECs proliferation.The effect of aPL on DVECs migration and angiogene-sis was observed by cell scratch assay and angiogenesis assay.Real-time PCR(RT-PCR)was used to detect the effect of aPL on MMP-2 and VEGF mRNA expression in DVECs.Western blot was used to detect the effect of aPL on ERK phosphorylation,VEGF and MMP-2 protein ex-pression in DVECs.Results:aPL inhibited the proliferation,migration and angiogenesis of DVECs.aPL can inhibit ERK phosphorylation,VEGF and MMP-2 expression in DVECs.The expression levels of p-ERK,VEGF and MMP-2 in DVEC cells were significantly increased after the addition of ERK agonists.Conclusion:aPL can reduce the expressions of VEGF and MMP-2 in DVECs by regulating ERK pathway,and then inhibit the proliferation,migration and angio-genesis of DVECs.
