摘要

Abstract

[Objective]To assess the effect of Lactobacillus plantarum(LP37)on growth,immunity,digestive activities and gut microbiota composition of Sebastes schlegelii.[Method]1 200 fish with a body weight of(32.23+9.38)g were selected and randomly divided into four groups(three parallels each group),designated as Control group,LP37 group,FF group,FF-LP37 group for a 45-day experiment.The control group was fed with basic feed,the LP37 group was mixed with 107 CFU/g of LP37 in the basic feed,the FF group was mixed with 20 mg/kg of Florfenicol(FF)in the basic feed,and the FF-LP37 group was first mixed with 20 mg/kg of FF,second with 107 CFU/g of LP37 in the basic feed.On the 30th day of the experiment,serum,liver and gut contents of fish in each group were collected to analyse growth,immunity,digestive activities and gut microbiota composition.[Result](1)The weight gain rate(WGR)and specific growth rate(SGR)of fish in each experimental group were significantly increased(P<0.05)compared with control group and the FF-LP37 group was the best;activities of protease(PPS)and α-amylase(AMS)in the serum of three experiment groups were significantly increased,while the activity of lipase(LPS)decreased(P<0.05).The activity of PPS in the intestinal contents increased,and there was a significant difference between LP37 group and FF-LP37 group(P<0.05).The activity of AMS increased,with only there was a significant difference in LP37 group(P<0.05).(2)Compared with control group,the activities of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in the serum and liver of LP37 group were significantly decreased(P<0.05),while activities of alkaline phosphatase(AKP)and superoxide dismutase(SOD)were increased(P<0.05),and the content of malondialdehyde(MDA)was reduced(P<0.05).The AST activity decreased but the ALT activity increased in FF group,while activities of AKP and SOD were not significant different(P>0.05),and the MDA increased(P<0.05).Compared with control group,the activities of ALT and AST in the serum and intestinal contents of FF-LP37 group were decreased(P<0.05),SOD was increased(P<0.05),and MDA was not significant different(P>0.05).(3)The results of microbial high-throughput sequencing showed that compared with control group,the diversity,richness and the OTUs of the gut microbiota were the highest in FF-LP37 group,while the diversity of the FF group increased.Compared to control group,the relative abundances of Proteobacteria,Firmicutes and Bacteroidetes increased in LP37 group,and the abundances of Lactobacillus and Lactococcus significantly increased;the relative abundance of Firmicutes increased in FF group,while the abundances of Lactobacillus decreased;the relative abundance of Proteobacteria increased in FF-LP37 group,while the relative abundance of Lactobacillus increased.The abundances of Pseudomonas and Vibrio in LP37 group,FF group and FF-LP37 group showed a decrease.These results were consistent with the Bugbase prediction analysis results.(4)Functional prediction analysis showed that L.plantarum and florfenicol did not have a significant impact on the gut microbiota function of S.schlegeli,mainly manifested in metabolic function.[Conclusion]In summary,LP37 has a beneficial effect on the cultivation of S.schleskii.LP37 can play a positive role in promoting health of fish.
