目的:探究半夏泻心汤对人结肠癌细胞系SW480 细胞上皮-间质转化的作用机制.方法:30 只SD大鼠随机分为半夏泻心汤组和空白组,每组 15 只,连续 7d 分别每日灌胃半夏泻心汤及蒸馏水(24.68 g·kg-1),腹主动脉采血离心制备半夏泻心汤含药血清及空白血清.以结肠癌细胞SW480 为研究对象进行体外实验,将细胞分为对照组、不同浓度的空白血清组及含药血清组.CCK-8 法检测SW480 细胞的存活率筛选最佳药物干预浓度与时间.细胞划痕实验检测细胞迁移能力.实时荧光定量PCR(RT-qPCR)和蛋白免疫印迹(WB)法检测各组细胞中上皮间质转化蛋白[E-钙黏蛋白(E-cadherin)、波形蛋白(Vimentin)],TGF-β1/Smads通路[转化生长因子-β1(TGF-β1)、Smads家族蛋白3(Smad3)、Smads家族蛋白7(Smad7)]mRNA及蛋白表达水平.结果:与对照组及10%空白血清组比较,20%半夏泻心汤含药血清干预24h能够显著抑制SW480 细胞增殖(P<0.05,P<0.01),降低细胞迁移能力(P<0.01),降低Vimentin、TGF-β1、Smad3 mRNA及蛋白表达水平,同时提高E-cadherin mRNA及蛋白表达水平(P<0.05,P<0.01),并提高Smad7 mRNA表达水平(P<0.01).结论:半夏泻心汤能抑制结肠癌细胞SW480 上皮-间质转化,其作用机制可能与抑制TGF-β1/Smads通路激活有关.

Objective:To explore the mechanism of pinellia heart-draining decoction(PHDD)on epithelial-mesenchymal transition of human colon cancer cell lines SW480.Methods:Thirty SD rats were randomly divided into PHDD group and blank group,with 15 rats in each group.PHDD and distilled water(24.68 g·kg-1)were administered daily for 7 consecutive days.Abdominal aortic blood was centrifuged to prepare PHDD-containing serum and blank serum.Colon cancer cell lines SW480 were used as the research object for this in vitro study.The cells were divided into control group,different concentrations of blank serum groups,and drug-containing serum groups.CCK-8 method was used to detect the survival rate of SW480 cells to screen the optimal drug intervention concentration and time.Cell migration was detected by scratch assay.Real-time fluorescence quantitative PCR(RT-qPCR)and Western blotting(WB)were used to detect the mRNA and protein expression of epithelial-mesenchymal transition proteins[E-cadherin,Vimentin]and TGF-β1/Smads pathway[transforming growth factor-β1(TGF-β1),Smads family protein 3(Smad3),Smads family protein 7(Smad7)]in each group.Results:Compared with the control group and the blank serum containing 10%group,the serum containing 20%PHDD could significantly inhibit the proliferation of SW480 cells(P<0.05,P<0.01),weaken the cell migration ability(P<0.01),reduce the expression of Vimentin,TGF-β1,Smad3 mRNA and protein,and increase the expression of E-cadherin mRNA and protein(P<0.05 or P<0.01),and increase the expression of Smad7 mRNA(P<0.01).Conclusion:Pinellia heart-draining decoction can inhibit the epithelial-mesenchymal transition of colon cancer cell SW480,and its mechanism may be involved in inhibiting the activation of TGF-β1/Smads pathway.
