益髓破血方对脑出血小鼠肠道菌群和PI3K/Akt信号通路调节的作用机制研究
Mechanism of Yisui Poxue Formula on Regulating Intestinal Microbiota and PI3K/Akt Signaling Pathway in Mice with Cerebral Hemorrhage

Abstract
Objective:To investigate the effects of Yishui Poxue formula(YPF)on the regulation of PI3K/Akt signaling pathway and intestinal flora in cerebral hemorrhage mice.Methods:A total of 150 SPF grade C57BL/6J mice were randomly divided into sham operation group,model group and traditional Chinese medicine group,with 48 mice in each group.The intracerebral hemorrhage mouse model was reproduced by autologous blood injection in the model group and traditional Chinese medicine group,and the sham operation group was only inserted with micro injection needle for several minutes without blood injection.Samples were taken on days 1,3,and 7,the neural function scores of the mice were observed,the brain water content was observed by dry and wet weight method,the brain histopathological changes were observed by HE staining,the contents of TNF-α,IL-4 and IL-18 were detected by ELISA,and the expression levels of PI3K,Akt,p-PI3K and p-Akt were detected by Western blot.The changes of intestinal flora diversity and species in each group were observed using 16s rDNA gene sequencing.Results:Compared with the model group,the traditional Chinese medicine group showed a decrease in neurological function scores and brain water content at days 3 and 7(P<0.05),a decrease in degenerated neurons(P<0.05),the concentration decrease of TNF-αand IL-18(P<0.05),the concentration increase of IL-4(P<0.05),and decrease of expression levels of p-PI3K and p-Akt,and these changes are more significant at day 7 than at day 3(P<0.05).The diversity of microbial communities in the model group mice decreased,while the relative abundance of rumen coccidae increased.YPF could enrich the diversity of bacterial communities and increase the relative abundance of lactobacilli.Conclusion:Yishui Poxue formula can improve the secondary injury of cerebral hemorrhage by increasing intestinal beneficial bacteria,reducing pathogenic bacteria,enriching bacterial diversity,and regulating PI3K/Akt pathway.关键词
