目的:研究育阴灵加减方对早发性卵巢功能不全(POI)模型大鼠的影响及其可能作用机制.方法:35%D-半乳糖造模成功的 POI 模型子代雌性大鼠 40 只随机分为模型组,育阴灵加减方高(0.562 g/kg)、中(0.281 g/kg)、低(0.141 g/kg)剂量组和西药组,西药组给予戊酸雌二醇溶液0.120 mg/kg,第4 天加灌醋酸甲轻孕酮片溶液0.960 mg/kg,随后两药停用 1d,为 1 个疗程,灌胃 4 个疗程.未造模的8 只子代雌性大鼠作为空白组.观察大鼠体质量、卵巢及子宫系数、卵巢组织的病理变化、血清抗缪勒管激素(AMH)、抑制素B(INHB)、血管内皮生长因子(VEGF)水平,蛋白免疫印迹法及荧光定量PCR法测定卵巢组织中Hippo信号通路相关效应因子表达情况.结果:与模型组相比,育阴灵加减方高、中剂量组可以升高大鼠体质量、卵巢系数、子宫系数(P<0.05),可改善卵巢组织结构并升高大鼠血清AMH、INHB、VEGF水平(P<0.05),两组Hippo信号通路相关效应因子Mst1、Lats2、YAP1、p-YAP、Smad7 的表达量明显降低(P<0.05).结论:育阴灵加减方可能通过调控Hippo信号通路的表达改善POI模型大鼠的卵巢功能.

Objective:To study the effect of modified Yuyinling formula(YYLF)on POI model rats and its possible mechanism.Methods:The POI rat model was established by 35%D-galactose,40 female rats were randomly divided into model group,Chinese medicine high-dose(0.562 g/kg),medium-dose(0.281g/kg),and low-dose(0.141g/kg)groups,and Western medicine group.The Western medicine group was gavaged diethylstilbestrol solution at a dose of 0.120 mg/kg,and on the fourth day,mifepristone solution at a dose of 0.960 mg/kg was gavaged,and then,both drugs were discontinued for 1 day,as a treatment course,and four treatment courses were carried out.Eight non-modeled female rats were used as the blank group.Observations were made on the body weight of rats,ovarian and uterine coefficients,pathological changes in ovarian tissue,serum levels of anti-Müllerian hormone(AMH),inhibin B(INHB),and vascular endothelial growth factor(VEGF).The expression of Hippo signaling pathway-related effector factors in ovarian tissue were detected using protein immunoblotting and fluorescence quantitative PCR.Results:Compared with model group,body weight,ovarian coefficient and uterus coefficient of rats in high and medium dose group of YYLF were raised(P<0.05);YYLF could improve ovarian tissue structure,could raise the serum AMH,INHB and VEGF levels(P<0.05),and could reduce the expression levels of Hippo signaling pathway related effect factors Mst1,Lats2,YAP1,p-YAP and Smad7.Conclusion:Modified Yuyinling formula may improve the ovarian function of POI model rats by regulating the expression of Hippo signaling pathway.
