FOXO3a在子宫内膜样癌中的表达及意义
Expression and significance of FOXO3a in endometrial carcinoma

Abstract
Objective To investigate the expression characteristics of Forkhead box O transcription factor 3a(FOXO3a)in endometrioid carcinoma,analyze its expression differences in different clinical and pathological features,and explore its value in predicting prognosis.Methods A total of 48 patients diagnosed with endometrioid carcinoma who underwent surgical resection at the Affiliated Hospital of North China University of Science and Technology from Janu-ary 2016 to May 2018 were selected as the observation group.Additionally,32 patients diagnosed with endometrial simple hyperplasia and 22 patients diagnosed with proliferative phase endometrium during the same period were selected as the control and normal control groups,respectively.The protein expression level of FOXO3a in the three groups was detected by immunohistochemistry(IHC).The relationship between FOXO3a expression and postoperative survival time was ana-lyzed using Kaplan-Meier analysis.Moreover,The Cancer Genome Atlas(TCGA)database was used to perform bioin-formatics analysis to observe the differences in FOXO3a mRNA expression between endometrioid carcinoma tissue and nor-mal tissue and its correlation with clinical pathological features.The UCSC XENA tool was used to analyze the correlation between FOXO3a mRNA expression level and overall survival.Results The positive expression rate of FOXO3a in endo-metrioid carcinoma tissue was significantly lower than that in the endometrial simple hyperplasia and proliferative phase en-dometrial tissue groups(14.58％,75％,and 95.5％,respectively).The difference was statistically significant(P=0.000).FOXO3a expression in endometrioid carcinoma showed no significant differences in age,differentiation degree,presence of tumor nodules/lymph node metastasis,depth of invasion,proliferative index,or mismatch repair deficiency(all P＞0.05).Survival analysis showed no correlation between FOXO3a protein expression level and overall survival time after surgery(x2=0.252 6,P=0.615 2).Bioinformatics analysis showed that FOXO3a mRNA expression in endo-metrioid carcinoma tissue was significantly decreased(P＜0.01)and significantly correlated with age(P=0.044),but not with histological grade,clinical stage,or overall survival(all P＞0.05).Conclusion FOXO3a expression is down-regulated in endometrioid carcinoma,but its expression level is not significantly correlated with prognosis.FOXO3a may be involved in the pathogenesis of endometrioid carcinoma,but its predictive value for prognosis is limited.关键词
